Vascular responses to leukotriene B4, C4 and D4 following FPL 55712, indomethacin, saralasin, phentolamine and verapamil in the conscious rat

Abstract: The pressor and vascular permeability effects of leukotrienes B4 (LTB4), C4 and D4 were investigated in conscious unrestrained rats. 2 Leukotrienes C4 and D4 (3.2-51 nmol kg-' i.v.) caused an acute dose-dependent elevation of the mean arterial pressure, which was maximal after 2 min and returned to control levels within 14 min. Heart rate was significantly reduced by the higher doses of LTC4 and LTD4. LTB4 (up to a dose of 51 nmol kg-') was essentially inactive. 3 These effects of LTC4 and LTD4 were abolished … Show more

“…At present it is not known whether FPL 55712 or YM 16638 can be used to distinguish between LTC4 and LTD4 receptors. Indeed, FPL 55712 has been described as a selective LTC4 (Bach et al, 1979) or LTD4 receptor antagonist (Casey et al, 1983), while others have failed to find a selective antagonism (Letts & Piper, 1982;Filep et al, 1987). YM 16638 was reported to be a 3 to 6 times more potent antagonist of LTC4 than LTD4 (Tomioka et al, 1988).…”

“…The experiments were performed on conscious, chronically catheterized male Wistar rats weighing 225-290 g. The animals were kept in individual metabolic cages and were prepared as described previously (Filep et al, 1987 (Rawson, 1943).…”

“…Constant, Quebec, Canada). The animals were housed in individual metabolic cages'and were prepared as described previously (Filep et al, 1987). Briefly, under anaesthesia (ketamine, 75 mg kg' and sodium pentobarbitone, 15 mg kg-') catheters were implanted into the abdominal aorta and vena cava through the central tail artery and left femoral vein, respectively.…”
