“…Leaf and stem samples were washed with running water thoroughly and approximately, 4 × 7 mm sized small tissue segments cut from active tissue margins of lesions. Small symptomatic pieces of samples taken from leaves and stems were surface sterilized by 1% sodium hypochlorite solution (laundry bleach) for one minute, 70% alcohol for one minute and rinsed in sterile distilled water three times of one minute each (Geiser et al, 2005;Summerell et al, 2006) and the prepared samples were macerated in sterile distilled water and crashed using laboratory mortar and pestle. Finally, the filtrates were diluted using sterile distilled water and wait for 30 min and allowed the bacteria to float and release from the tissues and then each suspension of the plates were streaked with sterile wire loop on the agar media.…”
