Vasopressin and platelets: a concise review

Abstract: Arginine vasopressin (AVP or vasopressin) is a neurophypophyseal peptide hormone with both vasopressor and antidiuretic properties. This review will briefly discuss the chemistry and physiology of AVP, with the main focus being on AVP and the hemostatic system. Recent data on the effects of AVP, and its analogues, on platelet function will be highlighted. AVP-induced platelet activation could explain, in part, the benefit seen with AVP infusions in gastrointestinal bleeding. These data also suggest that AVP ma… Show more

“…(t-PA) from endothelium in a dose-dependent manner [15,16] . While in hemophilia A and VWD positive eff ects of DDAVP are explained by the transient increase of FVIII and VWF, its well known effi ciency in some forms of hereditary and acquired thrombocytopathies is still a matter of debate [23] . It is assumed that the DDAVP-induced release of high molecular weight multimers of VWF which favours platelet adhesion and aggregation is only one of the possible mechanisms [18] .…”

“…Arginine vasopressin (AVP) which is absent or deficient in central diabetes insipidus, cannot be used as a reliable biomarker because of its short in vivo half-life 2 and its instability in isolated plasma. 3 Moreover, the large volumes of plasma required for radioimmunoassay analysis and the complexity and length (>24 h) of the analyses 4 make AVP difficult to assess routinely paediatrics. Copeptin, a peptide hormone released with AVP in equimolar amounts, 5 is more stable, and concentration measurements are more reliable.…”

“…It has been proven to reduce the bleeding tendency in many congenital and acquired disorders of platelet function [29]. While there is little doubt that the DDAVP effect in hemophilia and vWD results from increase of factor VIII and VWF in blood, its mechanism of action in platelet disorders is still a matter of debate [30].…”
