Eplingiella fruticosa (Salzm. ex Benth.) Harley & J.F.B. Pastore, also known as â€œalecrim de vaqueiroâ€� or â€œcowboy rosemaryâ€�, is a woody, perennial, and aromatic shrub that growsin the states of northeast Brazil, with the exception of Alagoas, MaranhÃ£o, and PiauÃ. The plant is widely used in this region and has analgesic, vasodilator, anti-inflammatory, and larvicidal properties.The objective of this study was to evaluate the effect of different substrates and indolebutyric acid (IBA) on cuttings of E. fruticosa for identifying effective practices for the vegetative propagation of this species. Apical cuttings with a standardized length of 10cm and two pairs of leaves were harvested from mother plants in the vegetative stage. Two experiments were conducted: the first experiment tested the effect of three substrates (commercial, commercial + vermiculite, and commercial + vermiculite + humus) and the second experiment evaluated five concentrations of IBA (0.0, 1.0, 2.0, 3.0, and 4.0gLâˆ’1) and three growth periods (30, 45, and 60 days). The experimental design was randomized blocks with four replications. The analyzed variables were survival of cuttings, number of rooted cuttings, root length per cutting (RLC), number of buds per cutting (NBC), leaf dry mass (LDM), root dry mass (RDM), and total dry mass (TDM). In thefirst experiment,there were significant differences in RLC, NBC, LDM,RDM, and TDM, with a better growth using the commercial substrate. In the second experiment, there was no interaction between the evaluated parameters. However, there was a positive associated effect of IBA addition and the growth period on RLC, NBC, LDM, and RDM. The highest predicted value was attained for a concentration of 1.5g Lâˆ’1IBA and a growth period of 60 days, for the above-mentioned variables.

Aging is associated with the development of various chronic diseases, in which both cardiovascular disorders and osteoarthritis are dominant. Currently, there is no effective treatment for osteoarthritis, whereas hypertension is often treated with L-type voltage-operated calcium channel blocking drugs, nifedipine being among the most classical ones. Although nifedipine together with other L-type voltage-operated calcium channel inhibitors plays an important role in controlling hypertension, there are unresolved questions concerning its possible effect on cartilage tissue homeostasis and the development of osteoarthritis. The aim of this study was to analyse the effects of nifedipine on metabolic processes in human chondrocytes and bone marrow mesenchymal stem cells. To better understand whether the metabolic effects are mediated specifically through L-type voltage-operated calcium channel, effects of the agonist BayK8644 were analyzed in parallel. Nifedipine downregulated and mitochondrial respiration and ATP production in both cell types. Analysis of cartilage explants by electron microscopy also suggested that a small number of chondrocyte mitochondria's lose their activity in response to nifedipine. Conversely, nifedipine enhanced glycolytic capacity in chondrocytes, suggesting that these cells have the capacity to switch from oxidative phosphorylation to glycolysis and alter their metabolic activity in response to L-type voltage-operated calcium channel inhibition. Such a metabolic switch was not observed in bone marrow mesenchymal stem cells. Nitric oxide activity was upregulated by nifedipine in bone marrow mesenchymal stem cells and particularly in chondrocytes, implying its involvement in the effects of nifedipine on metabolism in both tested cell types. Furthermore, stimulation with nifedipine resulted in elevated production of collagen type II and glycosaminoglycans in micromass cultures under chondrogenic conditions. Taken together, we conclude that the antihypertensive drug nifedipine inhibits mitochondrial respiration in both chondrocytes and bone marrow mesenchymal stem cells and that these effects may be associated with the increased nitric oxide accumulation and pro-inflammatory activity. Nifedipine had positive effects on the production of collagen type II and proteoglycans in both cell types, implying potentially beneficial anabolic responses in articular cartilage. These results highlight a potential link between antihypertensive drugs and cartilage health.

The present study evaluated the population structure and genetic diversity using ISSR molecular markers in 18 natural populations belonging to three species of Eplingiella (E. cuniloides, E. fruticosa and E. brightoniae), found growing naturally in the semiarid region of Northeast Brazil. Samples of 265 plants were analyzed using nine primer combinations, which generated 131 informative bands. Eplingiella spp. populations showed moderate genetic diversity (percentage of polymorphic bands, PPB = 75.6–96.9%, Nei's genetic diversity He = 0.31–0.39, Shannon's information index I = 0.33–0.48). Molecular variance analysis (AMOVA) revealed that within populations, variations contributed more (74%) to the genetic diversity than between population variations (26%), with percentage of the genetic differentiation coefficient (GST= 0.29). The mean value of FST was 0.175, demonstrating good differentiation between populations. The analysis of the structure by the Bayesian method revealed the formation of two groups (K = 2), with many migrant individuals and a high level of miscegenation. The hierarchical cluster dendrogram grouped the 18 populations into two major clusters, with good support for the main clades (100%). According to principal component analysis (PCA), the two main principal components explained 21.06% of the total variation. The ISSR molecular markers used were effective in identifying the variability of natural populations of Eplingiella spp., and population structure demonstrated recent diversification of species. The results shed more light on the genetic variation and evolutionary dynamics of Eplingiella, helping to formulate effective breeding strategies.
