Ventricular Myocyte Caspases Are Directly Responsible for Endotoxin-Induced Cardiac Dysfunction

Abstract: Background-Although most of the deleterious effects of sepsis-induced apoptosis have been attributed to increased lymphocyte cell death, caspase activation may directly alter cell function of different organ systems. We postulated that left ventricular (LV) cardiomyocyte caspase activation is directly involved in sepsis-induced heart contractile dysfunction. Methods and Results-LV cardiomyocytes isolated 4 hours after rat treatment with endotoxin injection (10 mg/kg) displayed major reductions in contractile r… Show more

“…Numerous studies have shown that apoptosis plays a significant role in the morbidity and mortality associated with sepsis (4). By way of example, prevention of apoptosis with caspase inhibitors significantly improved survival in murine CLP-induced sepsis (10,24). Support for this concept can also be found in the work of Bommhardt et al (4).…”

“…Cardiac myocyte apoptosis plays a major role in cardiac dysfunction following myocardial I/R injury (32)(33)(34). Activation of the PI3K/Akt signaling pathway decreases cardiac myocyte apoptosis following myocardial I/R (26 -29).…”

“…Activation of various caspases, the effectors of apoptosis, and mitochondrial cytochrome c release have been reported in cardiomyocytes following septic challenge (139)(140)(141). Caspase 3 activation via endotoxin might also be associated with altered calcium myofilament responses, cleavage of contractile proteins, and sarcomere disorganization (142). Therefore, it is not surprising that anti-apoptotic strategies have reversed cardiac dysfunction (inhibition of caspases [particularly caspase 3] averted endotoxin-induced cardiac dysfunction and heart apoptosis) (137,143).…”
