Vi4-miR-185-5p-Igfbp3 Network Protects the Brain From Neonatal Hypoxic Ischemic Injury via Promoting Neuron Survival and Suppressing the Cell Apoptosis

Abstract: Neonatal hypoxic ischemic encephalopathy (HIE) due to birth asphyxia is common and causes severe neurological deficits, without any effective therapies currently available. Neuronal death is an important driving factors of neurological disorders after HIE, but the regulatory mechanisms are still uncertain. Long non-coding RNA (lncRNA) or ceRNA network act as a significant regulator in neuroregeneration and neuronal apoptosis, thus owning a great potential as therapeutic targets in HIE. Here, we found a new lnc… Show more

“…In the Model group, rats were intraperitoneally injected with 3% of isoflurane before surgery. Referring to the previous method [27], the right common carotid artery of each rat pup was permanently ligated, and the pups were placed in an airtight chamber maintaining hypoxia (at 37°C with 92% of N 2 and 8% of O 2 at 4 L/min) for 2 h. In the Sham group, the common carotid artery of each rat pup was exposed as previously described [27], but without ligation and hypoxia. In the Model + DEX group, SD rat pups were treated as those in the Model group, and then rats were treated with 100 μL of 0.1 mg/kg DEX intravenously, once a day.…”

“…IGFBP3 activates the XBP1/IGFBP3/MMP-9 pathways to regulate the invasion and metastasis of non-small cell lung cancer (NSCLC) cells [ 36 ]. Studies have shown that Vi4 and miRNA-185-5p competitively combine with IGFBP3 to promote neuronal cell proliferation and reduce the risk of neonatal hypoxic ischemic encephalopathy (HIE) [ 37 ]. In view of the fact that no research related to muscles has been carried out, we made a bold attempt to deeply explore the effect of IGFBP3 in skeletal muscle cells of Hu sheep.…”

“…In brief, 3-day-old rat pups were fasted for 8 h before the experiment. Rats were subjected to the sustained inhaling anesthetization with 3% isoflurane [19]. The concentrated virus was injected into the ipsilateral (right side) ventricle (coordinates: 1.0 mm right to bregma and 1.5 mm to the lambdoid suture).…”
