To explore the influence of pH values on the properties of a compound system containing tea polyphenols (TPs) and low acyl gellan gum (LGG), the color, texture characteristics, rheological properties, water holding capacity (WHC), and microstructure of the compound system were measured. The results showed that the pH value noticeably affects the color and WHC of compound gels. Gels from pH 3 to 5 were yellow, gels from pH 6 to 7 were light brown, and gels from pH 8 to 9 were dark brown. The hardness decreased and the springiness increased with an increase in pH. The steady shear results showed that the viscosity of the compound gel solutions with different pH values decreased with increasing shear rates, indicating that all of the compound gel solutions were pseudoplastic fluids. The dynamic frequency results showed that the G′ and G″ of the compound gel solutions gradually decreased with increasing pH and that G′ was higher than G″. No phase transition occurred in the gel state under heating or cooling conditions at pH 3, indicating that the pH 3 compound gel solution was elastic. The WHC of the pH 3 compound gel was only 79.97% but the WHC of compound gels pH 6 and pH 7 was almost 100%. The network structure of the gels was dense and stable under acidic conditions. The electrostatic repulsion between the carboxyl groups was shielded by H+ with increasing acidity. The three-dimensional network structure was easily formed by an increase in the interactions of the hydrogen bonds.

Background: Diabetes is an endocrine disorder characterized by an abnormal increase in blood sugar level. Vildagliptin is a Dipeptidyl Peptidase-4 (DPP-4) inhibitor used in the treatment of type 2 diabetes mellitus. Purpose: The present work deals with the Quality by Design (QbD) approach based formulation and optimization of Vildagliptin-Eudragit RL 100 coated nanoparticles (VLD-EudNPs). Materials and Methods: VLD-Eud NPs were developed by combined technique of solvent evaporation and nano-precipitation. Optimization was done by 3 2 factorial designs. Prepared nanoformulations were characterized for percentage yield and morphology. Further in vitro drug release and kinetics were studied. The accelerated stability study of the optimized formulation was carried out for 3 months. Results: VLD-EudNPs ranged from 58.5±2.02 nm to 425.4±4.71 nm with a PDI of 0.27±0.036 to 0.52±0.086. Entrapment efficiency (% EE) of nanoformulations F1 to F9 ranged between 32.29±1.16 to 75.26±1.63%. The particles were found to be cylindrical-shaped, with a percentage yield of 54.7±3.45 to 84.4±1.51%. During the in vitro drug release study, at acidic pH, <5% drug release, while delayed release at basic pH, at the end of 24 hr, all formulations have released >68% of the drug following zero order kinetics. The optimized formulation has proven its stability during stability study. Conclusion: The present work concludes that prepared and optimized VLD-Eud NPs could be a better approach for sustained delivery of Vildagliptin for the management of diabetes mellitus as compared to traditional drug delivery.

Hydrogels have a tridimensional structure. They have the ability to absorb a significant amount of water or other natural or simulated fluids that cause their swelling albeit without losing their structure. Their properties can be exploited for encapsulation and modified targeted drug release. Among the numerous natural polymers suitable for obtaining hydrogels, gellan gum is one gaining much interest. It is a gelling agent with many unique features, and furthermore, it is non-toxic, biocompatible, and biodegradable. Its ability to react with oppositely charged molecules results in the forming of structured physical materials (films, beads, hydrogels, nanoparticles). The properties of obtained hydrogels can be modified by chemical crosslinking, which improves the three-dimensional structure of the gellan hydrogel. In the current review, an overview of gellan gum hydrogels and their properties will be presented as well as the mechanisms of ionotropic gelation or chemical crosslinking. Methods of producing gellan hydrogels and their possible applications related to improved release, bioavailability, and therapeutic activity were described.
