Vincristine: Beyond on anticancer treatment

Abstract: Vincristine is a chemotherapeutic agent that is a component of many combination regimens for a variety of malignancies, including several common pediatric tumors.78% of patients with advanced malignant disease were treated with vincristine, an alkaloid derived from vinca rosea Linn, 59% of these survived from the beginning of treatment and could be evaluated. Favourable responses were seen in patients with Hodking's disease, reticulum cell sarcoma, lymphosarcoma, carcinoma of the breast, acute leukemia and cho… Show more

“…Crude methanolic extracts of vincristine in combination therapy with conferone from Ferula schtschurowskiana increased its cytotoxicity in the cancer cells. Conferone competitively binds to P-gp transporters, decreases the vincristine resistance, thus, and enhances vincristine cytotoxicity [ 119 , 120 ]. In-vitro and in vivo clinical studies of vincristine sulphate demonstrated an increase in cell death when administered intraperitoneally.…”

“…1,2 However, the oculo-visual system has a potentially high sensitivity to toxic substances. 3,4 In this way, understanding ocular complications will help veterinarians and oncologists recognize them early and intervene before blindness occurs. Anticipating various treatment-related toxicities may allow pharmacists to develop intervention strategies that could minimize or eliminate an expected side effect.…”

“…Neurotoxicity is a common and dose-limiting adverse effect of vincristine in medical medicine, which usually occurs as cranial nerve palsy, and optic neuropathy/atrophy. 4,10 Depolarization of neurotubules by vincristine results in neurofibrillary degeneration and impairment of axonal transport. Blindness is a rare central neurotoxicity of vincristine therapy, with 11 reported cases in humans.…”
