-The significance of Acute Bee Paralysis Virus (ABPV) for the overwintering capacity of honeybee colonies was studied under field conditions. A case-control study of samples collected in Germany from 2004 to 2006 was performed. Successfully wintering colonies (control) were compared with winter fatalities (cases), using a binary logistic regression model that focused on the pre-winter ABPV burden as the explanatory variable. To quantify the ABPV burden a SYBR Green based real time PCR protocol was developed. An ABPV-specific primer pair hybridising to the RNA-dependent RNA-polymerase region was used. The assay proved to be ABPV specific and reproducible. ABPV infection was more often observed in colonies that did not survive the winter. In addition, winter fatalities showed higher ABPV burden than surviving colonies. Our findings provide support for a strong and presumably causal relationship between winter mortality, Varroa destructor and ABPV.Apis mellifera /acute bee paralysis virus / real time PCR / relevance / viral load / winter mortality / Varroa destructor

ABSTRACT:A honeybee colony, part of an apiary of nine, showed abnormalities in brood pattern and was thus presented for study. A classic veterinary medicine approach has allowed the diagnosis of a severe case of sacbrood virus (SBV) confirmed by a high viral load in affected larvae. SBV is known to infect larvae of the honeybee (Apis mellifera), resulting in failure to pupate and ultimately death of infected larvae. Several contributing factors combined, among them the parasite Varroa destructor, have been identified in this particular affected colony to explain the clinical outbreak of the disease whereas, in the majority of cases, infected colonies remain asymptomatic. As no specific cure of honeybee viruses is available, the management of these contributing factors is essential, including feeding of colonies and control of the Varroa parasite. After implementation of management solutions, the colony rapidly recovered in six weeks, but did not recommence honey production and remained at higher risk of a winter collapse. An earlier control management would have been more effective: regular visits of the colonies by the beekeepers should be the rule in order to detect abnormalities and also to detect and eliminate as early as possible the combination of factors that contribute to the proliferation of the virus.

This study aimed to assess the possible relationship between the presence of Israeli acute paralysis virus (IAPV) of honeybees and disease symptoms development at the colony level, to describe the IAPV load in field colonies and to illustrate phylogenetic relationships between IAPV isolates in Andalusia (Spain). Presence and load of IAPV was studied in 96 colonies from all provinces in Andalusia. Epidemiological surveys were performed in all the colonies to assess their sanitary status. IAPV was found in 13.5% of the sampled colonies, and no association was observed between the presence of IAPV and disease symptoms at the colony level. An average IAPV load was established in 4.9•105 genome equivalent copies per bee. Phylogenetic analysis revealed that Andalusian isolates belong to a different lineage to a previously described isolate found in Valencia (2010). The results of this study will help us understand the epidemiology and effect of IAPV on Spanish colonies.
