Viriplanin A, a new anthracycline antibiotic of the nogalamycin group. II. The structure of a novel hydroxyamino sugar from reduced viriplanin A.

Abstract: Methyl 2,3,6-trideoxy-3-hydroxyamino-3-C-methyl-a-D-r^(9-hexopyranoside (2) and the corresponding amino sugar (4) were isolated from reduced viriplanin A by acidic methanolysis and esterified to the di-p-bromobenzoates (3 and 5), respectively. The absolute configuration of crystalline 3 was determined by X-ray analysis to be a-D. This result could be confirmed by oxidation of 2 to methyl a-D-decilonitroside (6) and from the CDspectra of 3 and 5. Thus, the nitrogen-containing sugars of viriplanin A and probably… Show more

“…26,27 It also exists in several structurally related natural products including arugomycin ( 20 ), 28,29 cororubicin ( 21 ), 30 and viriplanin A ( 22 ). 31,32 Interestingly, these anthracycline-type antibiotics all contain a bicyclo-aminosugar moiety that is attached to the aglycone via a C -glycosidic bond in addition to the typical O -glycosidic linkage.…”

“…The preparation of a dinitroso derivative of a vinylogous tetrathiafulvalene has been reported. 28 The compound (16) has vicinal -N=O groups, and the proton NMR spectrum suggests a symmetrical structure which can be rationalized by a stabilization of the two NO groups by S + ---O − interactions as shown in (17). The compound is brown in colour unlike other dinitroso compounds, and does not exhibit the characteristic absorption band in the red.…”
