The compilation and analysis of the patient’s genealogies is one of the methods of population genetics, which makes it possible to identify a predisposition to a particular oncological pathology. At present, it is relevant to prove the feasibility of developing and introducing into clinical practice a comprehensive method for diagnosing and preventing tumors based on data from genetic counseling, molecular biological research and modern artificial intelligence technologies. An information-analytical system is proposed that allows analyzing the patient’s data obtained during the consultation, with the possibility of supplementing them with information from the medical history and the results of the study. The proposed information system is able to analyze of the genealogy and give a preliminary conclusion about the risk of a tumor process in the patient’s family members, according to the algorithms of the morbidity accumulated in the region.

ВЗАИМОСВЯЗЬ ЭКСПРЕССИИ ПРОТЕИНОВ NF1, р53, bcl-2, рRB С ДНК/РНК ВИРУСОВ И МУТАЦИЯМИ ГЕНОВ BRCA 1/2 и hMSH2 ПРИ РАКЕ ПЕЧЕНИ Аннотация. Среди опухолей желудочно-кишечного тракта заболеваемость и смертность от первичного рака печени остается высокой. За последние 20 лет заболеваемость раком печени в Беларуси увеличилась на 45,7 %.Цель исследования -установить взаимосвязь экспрессии протеинов NF1, р53, bcl-2 и рRB с ДНК/РНК вирусов и мутациями генов BRCA 1/2 и hMSH2 при раке печени.Обследованы пациенты (n = 303) с морфологически установленным первичным раком печени, гепатитом и здоровые лица. Выполнено молекулярно-биологическое и иммунологическое исследование уровней антител к протеинам клеточного цикла NF1, р53, bcl-2, рRB, ДНК/РНК вирусов, выявлены мутации генов BRCA 1/2 и hMSH2. Уровни антител к р53, bcl-2, рRB и NF1 в сыворотке здоровых лиц отличались от аналогичных показателей в образцах крови и экстрактов ткани пациентов с гепатитом, раком печени и в образцах нативной ткани печени. Мутации при раке печени в опухолевой ткани составили 6,13 % (p = 0,004), в крови -0,85 %. Изменения гена hMSH2 в ткани установлены в 15,48 % случаев. В ткани опухоли выделены ДНК/РНК вируса простого герпеса 1-го и 2-го типа (56,1 %), вируса герпеса 6-го типа (24,4 %), цитомегаловируса (17,1 %), вируса гепатита С (17,1 %), вируса гепатита В (4,9 %), вируса папилломы человека (4,1 %), вируса Эпштейна-Барр (2,4 %), а также микст-персистирующих форм инфекций (26,8 %). При гепатите (С, В) и раке печени с носительством цитомегаловируса наблюдался рост эксрессии bcl-2, NF1 и pRB (p = 0,001). Наличие вируса Эпштейна-Барр приводило к активности белка p53 при раке печени (p = 0,003). Наличие протеинов р53, bcl-2, рRB и NF1 в ткани печени не зависело от возраста и пола пациентов. Предполагается, что по степени их влияния на функцию генов, с учетом присутствующей в геноме ДНК/РНК вирусов и мутаций (BRCA 1/2 и hMSH2), можно определить степень онкогенности вирусов. Превышение предсказанных для лиц возрастной группы 50,8 года концентраций р53, bcl-2, рRB и NF1 в крови свидетельствовало о риске развития/ наличия опухолевого процесса в печени.Ключевые слова: рак печени, гены BRCA 1/2 и hMSH2, вирусы, клеточный цикл, опухоль Для цитирования: Кузнецов, О. Increasing incidence, difficulties in early diagnosis, and a high mortality rate in liver cancer (LC) determine the relevance of studying the mechanisms of its development. The aim of the work is to evaluate the expression of high molecular weight glycoproteins MUC-1, MUC-13 in liver cancer. The object of study is LC tissue samples of 65 patients from the archives and 34 blood serum samples from patients with morphologically confirmed LC. The age of subjects was 26– 97 years. The level of antibodies to MUC-1 and MUC-13 was studied by ELISA. The reference value ranges of MUC-1 (0.250 ± 0.10 ng/ml) and MUC-13 (0.321 ± 0.13 ng/ml) in the blood serum of healthy individuals were established. The concentration of antibodies to MUC-1 and MUC-13 in the blood serum in RP was significantly higher than that in practically healthy individuals. The concentration of MUC-1 and MUC-13 in the LC tumor tissue was higher than that in the blood serum of apparently healthy individuals and LC patients. With a confirmed LC diagnosis, the level of antibodies to MUC-1 in the blood serum, which exceeds 0.373 ng/ml, and the level of antibodies to MUC-13, which is more than 0.939 ng/ml, may indicate a high risk of a tumor process.
