Potato is an economically important agro-industrial crop that is conventionally propagated, however; its potential transmission of viruses through seed tubers from generation to generation is a major limitation of potato yield production. In order to produce potato virus-free and sufficient amount of potato seed tubers, several approaches of in vitro methods for virus elimination have been developed. Meristem culture has been used alone or combined with techniques such as thermotherapy, electrotherapy, cryotherapy and chemotherapy as the best alternative method for treating potato infected by viruses. Recent literature has shown that to eliminate potato virus significantly depends upon the potato cultivar, antiviral agents, type of virus, the duration of heat treatment. Appropriate duration for efficiency elimination is still under investigation. Viral elimination rate can be detected through serological methods such as enzyme-linked immunosorbent assay (ELISA) and molecular biology technique such as real time reverse transcriptase polymerase chain reaction (real time RT-PCR) that are used for pre and post elimination virus detection to evaluate the efficiency and the accuracy of virus elimination method. The purpose of this review is to highlight virus elimination methods in potato and recommending the most effective tool for virus detection in order to ensure the production of potato plantlet free of viruses. Resumen La papa es un cultivo de importancia económica agro-industrial que se propaga de manera convencional, no obstante, la potencial transmisión de virus a través del tubérculo-semilla de generación en generación es una limitación mayor en la producción del rendimiento. Con el fin de producir papa libre de virus y suficiente cantidad de tubérculo-semilla de papa, se han desarrollado varias estrategias de métodos in vitro para la eliminación de los virus. El cultivo de meristemos se ha usado solo o combinado con técnicas tales como termoterapia, electroterapia, crioterapia y quimioterapia, como el mejor método alternativo para tratar papa infectada por virus. La literatura reciente ha demostrado que para eliminar virus significativamente se depende de la variedad de papa, agentes antivirales, tipo de virus, la duración del tratamiento térmico. Aun esta bajo investigación la duración apropiada para la eliminación eficiente. El nivel de la eliminación del virus puede detectarse por métodos serológicos, tales como el inmunoensayo con enzimas conjugadas (ELISA) y técnica de biología molecular, como la reverso-transcripción de reacción en cadena de la polimerasa en tiempo real (RT-PCR) que se usan para la detección del virus pre y post eliminación para evaluar la eficiencia y la precisión del método de eliminación del virus. El propósito de esta revisión es resaltar los métodos de eliminación de virus en papa y en la recomendación de la herramienta más efectiva para la detección de virus para asegurar la producción de plántulas de papa libres de virus.

Virus infection in potato reduces yield through seed degeneration. This can be reduced by use of virusfree seed tubers. However, novel approaches are required to ensure availability of virus-free stocks, especially in developing countries where the seed potato system are evolving. Consequently, a laboratory experiment was conducted at Kachwekano ZARDI in Uganda to evaluate and determine appropriate procedures for cleaning valuable potato cultivars infected with the most prevalent potato viruses; potato virus X (PVX) and potato virus S (PVS) which are frequent in the potato farming system. Thus, 20 in-vitro potato plantlets from cultivars Victoria, Kinigi and Rwangume each batch infected with either PVX or PVS in three replicates were grown in a thermotherapy chamber for two, three and four weeks at 37 to 40°C in 16 h of light and 30 to 34°C for 8 h of darkness per day. An equal number plantlets and replicates of the same cultivars and virus infection combinations were grown in a standard tissue culture (TC) growth room at 16 to 18°C with 16 h of light without thermotherapy as controls. Results indicated that plantlet survival after in-vitro thermotherapy decreased with increasing duration of heat treatment. Virus elimination efficiency significantly (P ≤ 0.05) differed between heattreated plantlets and the controls but not between the duration of thermotherapy treatment. However, the highest proportion of virus-free plantlets was obtained after three weeks of thermotherapy. Significant (P ≤ 0.05) interactions were observed between thermotherapy duration and virus type on virus elimination efficiency where significantly (P ≤ 0.05) more PVS-free than PVX-free plantlets were obtained at the same thermotherapy duration. Three weeks of thermotherapy of virus-infected in-vitro plantlets, particularly for PVS, offered an equilibrium duration for adequate plantlet survival and maximum meri-clone regeneration to obtain the highest proportion of virus-free plantlets. The use of thermotherapy to obtain a high proportion of PVX-free plantlets from infected potato seed stock showed no clear trend and needs further investigation.

ABSTRAKPenggunaan benih bebas virus merupakan salah satu cara pengendalian penyakit virus. Jaringan tanaman dapat dibebaskan dari virus melalui aplikasi teknik eliminasi virus, seperti termoterapi, kemoterapi, kultur meristem, dan krioterapi. Tujuan penelitian ini adalah untuk menguji respon varietas tebu terhadap perlakuan teknik kultur meristem dan kemoterapi dengan bahan antiviral, serta untuk mengetahui efektivitasnya dalam mengeliminasi virus sugarcane streak mosaic virus (SCSMV) pada tebu. Penelitian ini dilakukan pada April−November 2015 di Laboratorium Kultur Jaringan, Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumberdaya Genetik Pertanian dan Laboratorium Virologi, Fakultas Pertanian, Universitas Gadjah Mada. Penelitian terdiri atas tiga tahap, yaitu 1) Deteksi virus dari tanaman induk, 2) Aplikasi teknik kultur meristem dan kemoterapi, serta 3) Indeksing virus. Bahan tanaman yang digunakan adalah sebelas varietas tebu (GMP3, PS865, dan Kentung asal Bogor, PS862 dan Cening asal Cirebon, PS881 asal Jember, PSJK922 asal Malang, serta PS864, PS881, PSJK922, PSJT941 asal Pati). Deteksi virus dilakukan secara RT-PCR dengan primer universal MJ dan primer spesifik SCSMV. Bahan antiviral yang digunakan untuk kemoterapi adalah Ribavirin (0 dan 25 µg/l). Hasil uji RT-PCR menggunakan primer universal MJ menunjukkan bahwa empat varietas (GMP3 asal Bogor, PS864 dan PSJT941 asal Pati, serta Cening asal Cirebon) terinfeksi oleh Potyvirus. Empat varietas lainnya (PS862 asal Cirebon, PS881 asal Jember, PSJK922 asal Malang, dan Kentung asal Bogor) terbukti terserang virus SCSMV berdasarkan uji RT-PCR dengan primer spesifik. Seluruh meristem mampu beregenerasi membentuk tunas. Penggunaan Ribavirin 25 µg/l tidak menyebabkan penurunan daya tumbuh meristem (50−100%), bahkan seluruh varietas mampu bermultiplikasi tunasnya dibandingkan dengan kontrol yang hanya memiliki daya tumbuh 0−100%, dan tidak semua varietas mampu bermultiplikasi tunasnya. Secara tunggal, aplikasi teknik kultur meristem tidak mampu mengeliminasi virus SCSMV, namun jika dikombinasikan dengan perlakuan kemoterapi maka virus SCSMV dapat tereliminasi dengan efikasi sebesar 44,4%.Kata kunci: Tebu, Ribavirin, Potyvirus, SCSMV, RT-PCR ABSTRACTThe use of virus-free seedling is an option for controlling viral disease that can be obtained through the application of viral elimination method.
