Visual detection of tropomyosin, a major shrimp allergenic protein using gold nanoparticles (AuNPs)-assisted colorimetric aptasensor

Abstract: A gold nanoparticle-based label-free colorimetric assay was developed to detect the shrimp allergenic protein tropomyosin (TM), an important biomarker responsible for severe clinical reactivity to shellfish. In a gold nanoparticles (AuNPs)-tropomyosin-binding aptamer (TMBA) complex, the aptamer adsorbs onto the surface of AuNPs and dissociates in the presence of TM. In addition, AuNPs tend to aggregate in the presence of ionic salt, revealing a color change (i.e., wine-red to purple/ blue) with a shift in the … Show more

“…AuNPs-based aptasensors have been widely applied in the detection of various targets including chemical compounds, proteins, pesticides, toxins, and viruses [ 36 , 38 , 39 , 40 , 48 , 49 , 50 ]. Their simple preparation, rapid detection, and the fact that they do not require special instrumentation have made them the preferred choice for PoCT [ 51 ].…”

“…In addition, a gold nanoparticle‐based label‐free colorimetric assay was proposed as a visual colorimetric detection technology to detect the shrimp tropomyosin. The aptasensor is highly reliable, selective, and sensitive with the observed detection limitation of 70 nmol/L in shrimp (Pavase et al., 2021). However, it was reported that the aptamer stability and function might be altered due to the instability and relative nonspecific binding of aptamers (Stoltenburg et al., 2007).…”

It is worthy notable that these biomolecules facilitate the biosensing of analytes at concentrations as low as some femtograms per milliliter [86][87][88][89][90]. Note: *NPs = Nanoparticles…”
