Visualization of two distinct classes of neurons by gad2 and zic1 promoter/enhancer elements in the dorsal hindbrain of developing zebrafish reveals neuronal connectivity related to the auditory and lateral line systems

Abstract: The dorsal hindbrain includes distinct classes of neurons for processing various sensory stimuli, but the developmental aspects of these neurons remain largely unknown. We identify here two distinct classes of neurons in the dorsal hindbrain of developing zebrafish: (1) neurons that express the inhibitory neuronal marker Gad1/2, and (2) neurons that express the zn-5 antigen and Lhx2/9 and require the basic helix-loop-helix transcription factor Atoh1a for development. Neurons were traced to their axon terminals… Show more

“…In a divergent neural submap, each neuron forms ϳ60 boutons that concentrate a well characterized presyn- aptic marker. If we assume that each bouton represents a synaptic contact with one output neuron, the posterior lateral line has no more than 2,400 second-order targets in the zebrafish larva (Fame et al, 2006;Sassa et al, 2007). In a convergent submap, many lateralis neurons directly contact the lateral dendrite of the Mauthner cell.…”

“…As stated in the Introduction section, we became aware that the previous studies employing "GAL4VP16" in zebrafish (Davison et al, 2007;Inbal et al, 2006;Köster and Fraser, 2001;Sagasti et al, 2005;Sassa et al, 2007;Scott et al, 2007) actually used GAL4VP16 (Barlev et al, 1995) instead of the original GAL4VP16 construct (Sadowski et al, 1988). Direct give reporter expression…”

“…3). Toxicity has not been described in transgenic zebrafish lines that express GAL4VP16 413-470 under the control of tissue-specific regulatory elements from the genes isl1, gsc, gad2, and zic1 (Inbal et al, 2006;Sagasti et al, 2005;Sassa et al, 2007). Davison et al (2007) also used GAL4VP16 in their gene-trap constructs without observing deleterious effects.…”

“…Neurons that derive from the same neuroblast population could express a different combination of proteins depending on their position along a morphogen gradient, whose spatial distribution could vary over time as the brain enlarges. The generation of transgenic animals that label candidate second-order neurons in the MON will allow for testing of this possibility in vivo (Fame et al, 2006;Sassa et al, 2007;Scott and Baier, 2009). Regardless of the mechanism involved, our results provide a framework within which to assess the impact of timing in the progression of somatotopy.…”
