Vitamin D in health and disease: Current perspectives

Abstract: Despite the numerous reports of the association of vitamin D with a spectrum of development, disease treatment and health maintenance, vitamin D deficiency is common. Originating in part from the diet but with a key source resulting from transformation by exposure to sunshine, a great deal of the population suffers from vitamin D deficiency especially during winter months. It is linked to the treatment and pathogenesis and/or progression of several disorders including cancer, hypertension, multiple sclerosis, … Show more

“…Low level of vitamin D3 is associated with osteoporosis, rickets, cardiovascular disorder, some autoimmune disease, multiple sclerosis, diabetes mellitus and some cancers (9). It has been suggested that low level of vitamin D decreases the survival of general population and CKD and HD patients (10,11).…”

“…Others have demonstrated that diet has an important effect on vitamin D levels and this should not be overlooked. 8 However, since so many children do not have adequate vitamin D in their diets it should not be surprising that the prevalence of vitamin D insufficiency is high. 9,10 This finding, in combination with the recent increase in the recommended daily vitamin D intake for children suggests that far more of our patients than previously recognized require vitamin D supplementation.…”

“…The precursors of vitamin D are vitamins D 3 (cholecalciferol) and D 2 (ergocalciferol) [6]. Vitamin D 3 is formed by exposure to solar ultraviolet B (UVB) rays from its precursor, 7-dehydrocholesterol, present in the skin.…”

“…The 25(OH) D released by the liver is not the biologically active form of vitamin D, as that still needs a kidney hydroxylation to become an active compound. Action by the kidney turns 25(OH) D to 1,25 dihydroxyvitamin D (1,25(OH) 2 D) or calcitriol, the active form of vitamin D [6].…”
