VKORC1 Pharmacogenetics and Pharmacoproteomics in Patients on Warfarin Anticoagulant Therapy: Transthyretin Precursor as a Potential Biomarker

Abstract: BackgroundRecognizing specific protein changes in response to drug administration in humans has the potential for the development of personalized medicine. Such changes can be identified by pharmacoproteomics approach based on proteomic technologies. It can also be helpful in matching a particular target-based therapy to a particular marker in a subgroup of patients, in addition to the profile of genetic polymorphism. Warfarin is a commonly prescribed oral anticoagulant in patients with prosthetic valve diseas… Show more

“…Although there have been significant advances in the understanding of interindividual variability in response to warfarin, it is important to note that at least 40% of the variance in daily dose requirements remains unexplained, and is therefore a limitation of the reductionist approach. Other factors that may influence the response to warfarin such as microRNA and proteomics have been investigated, but further more global approaches that are integrated with the pharmacogenomic determinants are required.…”

“…Turning toward pharmaceutical toxicology, iTRAQ measurements of plasma were included with metabolomics methodologies in the study of drug toxicity in relation to the prevention and treatment of thromboembolic conditions . An iTRAQ based LC‐MS/MS pharmacoproteomics approach has also been used to analyze plasma protein profiles of 53 patients with a high ( n = 28) and low dose of warfarin therapy ( n = 25) …”
