Vps34 Inhibits Hepatocellular Carcinoma Invasion by Regulating Endosome-Lysosome Trafficking via Rab7-RILP and Rab11

Abstract: The role of Vps34, an indispensable protein required for cell vesicular trafficking, in the biological behavior of hepatocellular carcinoma (HCC) has yet to be studied. Materials and MethodsIn the present study, the expression of Vps34 in HCC and the effect of Vps34 on HCC cell invasion was detected both in vivo and in vitro. Furthermore, by modulating the RILP and Rab11, which regulate juxtanuclear lysosome aggregation and recycling endosome respectively, the underlying mechanism was investigated. ResultsVps3… Show more

“…In addition, inhibition of PIK3C3 reduced invasion and promoted the death of trametinib-resistant melanoma cells in an in vivo zebrafish xenograft of metastatic melanoma [112]. In contrast, PIK3C3 inhibits the invasion of HCC cells by regulating endosome-lysosome trafficking via the Rab7-RILP pathway [10]. Our study showed that inhibition of PIK3C3 by miR-338-5p suppresses autophagy and induces CRC cell EMT, migration, invasion, and metastasis [12] (Figure 3).…”

“…Published research on PIK3C3 expression in human cancer tissues is scarce. Both increased and decreased levels of PIK3C3 protein have been reported in HCC [10,106]. In a series of 31 pairs of normal and esophageal SCC tissues, PIK3C3 mRNA levels were significantly lower in esophageal SCC tissues [92].…”

“…The PI3K/AKT/mTOR signaling pathway is one of the most critical intracellular signaling pathways controlling basic cellular functions, including cell proliferation, survival, metabolism, and locomotion (Yu et al 2021 ; Corti et al 2019 ). Using RNA-seq data analysis, we uncovered the mechanism of action of RILP in osteosarcoma, at least in part, through its regulation of the PI3K/AKT/mTOR pathway, which in turn regulated the proliferation, migration, and invasion of osteosarcoma cells.…”

“… 14 Vps34 controls lysosomal transport to prevent the tumor cells invasion in HCC. 15 It has been noted that GNS561 alters lysosomal function to have anti-HCC effects. 16 Lysosomal TRPML1 controls tumor cell mitochondrial activity to encourage HCC growth.…”
