What about Platelet Function in Platelet Concentrates?

Abstract: The characterization of platelet concentrates (PCs) in transfusion medicine has been performed with different analytical methods and platelet lesions (from biochemistry to cell biology) have been documented. In routine quality assessment and validation of manufacturing processes of PCs for transfusion purposes, only basic parameters are monitored and the platelet functions are not included. However, PCs undergo several manipulations during the processing and the basic parameters do not provide sensitive analys… Show more

“…In our study, platelet inactivation on day 1 was likely due to "platelet storage lesion", a phenomenon resulting in the platelet's disc-to-sphere transformation and known to be associated with reduced responsiveness to adenosine diphosphate for platelet aggregation [22]. We believe our observation was due to hypoxia, an effect of the sample resting on the bench.…”

“…The comparability of pooled and apheresis platelet concentrates is a much-debated topic. 6 To avoid an influence of donor variability as well as of storage, 11 Note: Total blood volume was calculated after the Nadler formula. The pH analysis was performed at the end of storage from the manual transported platelet concentrate (PC).…”

“…The comparability of pooled and apheresis platelet concentrates is a much‐debated topic. 6 To avoid an influence of donor variability as well as of storage, 11 we used apheresis platelet concentrates on the day of donation. The strength of this study was that each concentrate presented its own control as each of it was split into three equal parts.…”

“…Further studies should be conducted to check if those biotinylation processes could be combined with different processes or storage conditions, such as Intercept treatment after the biotinylation, refrigeration, or cryopreserve BioPLTs storage 31 . All these parameters could be implemented in clinical trials to evaluate the in vivo function of transfused platelets and to investigate the impact of PC processing and storage conditions on the efficacy of transfusion 14 …”
