Wheezing preschool children with early‐onset asthma reveal a specific metabolomic profile

Abstract: Metabolomic urinary profile can discriminate preschoolers with recurrent wheezing who will outgrow their symptoms from those who have early-onset asthma. These results may pave the way to the characterization of early non-invasive biomarkers capable of predicting asthma development.

“…The urine samples were analyzed at the Mass Spectrometry Laboratory, Department of Women's and Children's Health, Padova University Hospital, Fondazione Istituto di Ricerca Pediatrica Città della Speranza. The sample preparation method and the chromatographic and mass spectrometry conditions were as described in a previous work [9,11] and elsewhere in this article (Supplementary Materials). Briefly, the metabolic profile of the urine was acquired using a Waters Acquity Ultra-Performance Liquid Chromatography system coupled to a Waters Q-TOF Synapt G2 mass spectrometer (Waters, Milford, MA).…”

“…Recently, an approach called "metabolomics" [6,7] has revealed the potential to identify the metabolic features of certain conditions, differentiating between phenotypes of the same disease and enabling a better characterization of the pathological mechanisms involved. Metabolomic studies are typically performed on biofluids, and urine samples are among the most often used in medical research, especially studies of children, because they can be collected using simple, noninvasive techniques [8][9][10].…”

“…56 Wheezing preschool children have a characteristic metabolic urinary profile that can be used to predict if they will develop earlyonset asthma after a 3-year follow-up. 57 Dimethylamine and allantoin are gut microbial-derived metabolites that have been associated with asthma development in early childhood and are potential prognostic markers of asthma. 58 A comprehensive systematic review and meta-analysis has recently reviewed bronchial hyperresponsiveness (BHR) in preterm-born subjects.…”

“…The role of high levels of metabolites such as butyrate for protection of allergy has been well characterized earlier in cohorts of farming children . On the other hand, various studies published recently in this journal have linked specific gut microbiota patterns with allergies . Also, indirect interventions, such as early‐life antibiotic use and its impact on the gut microbiome, are known to influence the development of allergy .…”
