Hordeum vulgare (Barley) and Panax ginseng have antioxidant activity referring to their diverse phytonutrient. Hypothyroidism in adult female rats was induced by pituitary-gonadal-adrenal disturbance, depleting the serum FSH levels with the elevation of corticosterone, prolactin, progesterone and testosterone hormones as well as (ERK1/2). Hypothyroidism evoked an oxidative stress status by increasing 8-hydroxy guanosine, which initiated apoptosis by uplifting apoptotic marker Caspase-3 both in serum and brain tissues. This is confirmed by the increase in the percentage of DNA-damage in the brain tissues. Significant decrease in all monoamines’ levels in different brain areas, downregulation of dopamine and 5-hydroxytryptamine receptors transcription, with a significant increase in excitatory amino acids was noted. Barley and ginseng renormalized cortisol and oxidative stress markers by increasing cellular resistance to stress and potentiated the role of the immune system through phytosterol and ginsenosides, so they considered potent free radical scavengers. Barley and Panax ginseng ameliorate the hormonal and neural dysfunction resulting from hypothyroidism, so they are recommended for relieving stress and improving mood and depression.

Previous studies have shown the therapeutic properties of ginseng and ginsenosides on hyperactive and impulsive behaviors in several psychiatric diseases. Herein, we investigated the effect of Panax ginseng Meyer (PG) on hyperactive/impulsive behaviors in a manic-like animal model, sleep deprivation (SD) rats. Male rats were sleep-deprived for 48 h, and PG (200 mg/kg) was administered for 4 days, from 2 days prior to the start of SD to the end date of SD. The elevated plus maze (EPM) test showed that PG alleviated the increased frequency of entries into and spent time within open arms by SD. In order to investigate the molecular mechanism on this effect of PG, we assessed differentially expressed genes (DEGs) in the prefrontal cortex of PG-treated SD rats using RNA sequencing (RNA-seq) and performed gene-enrichment analysis for DEGs. The gene-enrichment analysis showed that PG most prominently affected the glutamatergic synapse pathway. Among the glutamatergic synapse pathway genes, particularly, PG enhanced the expressions of glutamate transporter Slc1a3 and Slc1a2 reduced in SD rats. Moreover, we found that PG could inhibit the SD-induced phosphorylation of the NR2A subunit of the NMDA receptor. These results suggested that PG might have a therapeutic effect against the manic-like behaviors, regulating the glutamatergic neurotransmission.

Depression is closely linked to hypothalamus-pituitaryadrenal axis hyperactivity. Honokiol, a biphenolic lignan compound obtained from the traditional Chinese medicine Magnolia officinalis, can reduce the activity of the hypothalamus-pituitary-adrenal axis and improve depression-like behavior caused by hypothalamuspituitary-adrenal axis hyperactivity. The current study investigated the specific mechanism of action of this effect. A depression model was established by repeated injections of corticosterone to study the antidepressantlike effect of honokiol and its potential mechanism. Honokiol prevented the elevated activity of the hypothalamuspituitary-adrenal axis and the depression-like behavior induced by corticosterone. Treatment with honokiol resulted in greater glucocorticoid receptor mRNA expression, greater glucocorticoid receptor-positive expression, and a greater ratio of glucocorticoid receptor to the mineralocorticoid receptor in the hippocampus. Moreover, honokiol treatment led to lower levels of interleukin-1β in serum and the positive expression of the interleukin-1β receptor in the hippocampus. These results demonstrate that the antidepressant-like mechanism of honokiol, which has effects on inflammatory factors, may act through restoring the typical activity of the hypothalamus-pituitary-adrenal axis by regulating the glucocorticoid receptor-mediated negative feedback mechanism and the balance between glucocorticoid and mineralocorticoid receptors.
