Whole-Genome Sequences of Chlamydia psittaci Strain HJ, Isolated from Meat Pigeons with Severe Respiratory Distress and High Mortality

Abstract: The obligate intracellular Gram-negative bacterium Chlamydia psittaci causes systemic disease in psittacine birds, domestic poultry, and wild fowl. Importantly, C. psittaci may cause pneumonia, encephalitis, endocarditis, and even death in humans. The potential of pigeons as a source of human psittacosis is supported worldwide by relatively high seroconversion rates in the birds. This study reports the whole-genome sequencing of C. psittaci strain HJ, isolated from meat pigeons suffering from severe pneumonia … Show more

“…A highly virulent HJ strain of C. psittaci was isolated from diseased pigeons 11 while avian influenza virus (H9N2/chicken/Shandong/2011) 3 and Ornithobacterium rhinotracheale (ORT/chicken/Shandong/2011) 3 were isolated from the broilers with hemorrhagic pneumonia.…”

“…These pathogens should be considered when developing prevention strategies for respiratory disease in broilers 7 , 8 . Moreover, high seroprevalence of C. psittaci has been determined in Belgium, France and China with seropositive rates of 96%, 90% and 77.8%, respectively in broiler flocks 9 – 11 . A virulent C. psittaci strain has been isolated from meat pigeon farms with an outbreak involving severe respiratory signs 11 .…”

“…Low virulence strain CB3 and high virulence HJ strain of C. psittaci were isolated from free-living birds 27 and diseased pigeons 28 . AIV H9N2/chicken/Shandong/2011 was isolated from broilers as described previously 11 .…”

“…MLST provides higher discrimination and is robust, but requires time-consuming sequencing of seven gene targets. With the increase in number of whole genome sequences (WGS) available for C. psittaci strains [ 12 , 13 , 14 , 15 , 16 ], it is now possible to apply single nucleotide polymorphisms (SNPs) for the establishment of a phylogenetic tree pointing out true relationship between C. psittaci isolates. In our study, we aimed to develop a highly discriminative and user-friendly method where the work comprises three parts: (i) to identify clusters, based on SNP analysis, (ii) to compare those with MLST- and omp A-based topologies and (iii) to develop a rapid typing tool based on the identification of relevant SNPs by PCR/high-resolution melting (HRM) technology for straightforward genotyping of C. psittaci from isolates and clinical samples.…”
