Abstract

In order to clarify the synergism pathogenic mechanism of alfalfa mosaic virus (AMV) and white clover mosaic virus (WCMV) combined infection Nicotiana benthamianaunder indoor conditions, In this experiment, the effect of AMV and WCMV combined infection on the chlorophyll content, morphology and microstructure in N. benthamiana, the relative expression of the two viruses at different infected times were observed. The results showed that AMV and WCMV combined infection the chlorophyll content was significantly lower than that in N. benthamianaalone infection and healthy; electron microscopy observed the cytoplasmic ultralesions found that two forms of viruses in the AMV and WCMV combined infection of N. benthamiana, the length of bacillus virus was 20~75 nm observed in AMV alone infection, the length of linear virus was 400~600 nm observed in WCMV alone infection; after the AMV and WCMV combined infection of N. benthamiana, chloroplasts showed cleavage, osmium granules, mitochondrial crest voids increased, mitochondrial malformations were serioused, the chloroplasts internal layer structure was loose and flocculent in N. benthamiana by AMV、WCMV alone infection; real-time quantitative detection showed that the relative expression of AMV and WCMV reached the maximum on the 21 d, which increased by 39.46% and 31.53% compared with AMV and WCMV alone infection.
