Wnt/β-catenin signaling in kidney injury and repair: a double-edged sword

Abstract: The Wnt/β-catenin signaling cascade is an evolutionarily conserved, highly complex pathway that is known to be involved in kidney injury and repair after a wide variety of insults. While the kidney displays an impressive ability to repair and recover after injury, these repair mechanisms can be overwhelmed, leading to maladaptive responses and eventual development of chronic kidney disease (CKD). Emerging evidence demonstrates that Wnt/β-catenin signaling possesses dual roles in promoting repair/regeneration o… Show more

“…A variety of nephrotoxic events lead to marked accumulation of β-catenin in renal tubules [20,21]. In both folic acid nephropathy and ischemia-reperfusion, tubule-specific ablation of β-catenin is harmful, leading to increased mortality, renal injury, and tubular cell apoptosis [18].…”

“…For instance, besides Shh signaling pathway, TGF-β1, Wnt/β-catenin and Notch pathways are well recognized to play crucial roles in the evolution of renal fibrosis in CKD (Bielesz et al, 2010; He and Dai, 2015; He et al, 2009; Tan et al, 2014; Zhou et al, 2016). However, the role of Shh signaling is quite unique in this process in that it specifically targets interstitial cells, while it is mainly produced by the injured tubules.…”

“…Given the significant role of β-catenin in kidney development and repair after AKI [15, 20], we evaluated the expression of β-catenin in tubular cells after CM treatment. In the cisplatin group, the protein expression of β-catenin was significantly lower than that in the control group.…”

“…Wnt signaling, one of the evolutionarily conserved pathways, is associated with organ development, homeostasis and tissue repair [13-15]. When wnt ligands bind to Frizzled receptors and LRP5/6 coreceptors, the β-catenin degradation complex is sequestered, leading to the translocation of β-catenin to the nucleus to prompt the expression of target genes regulating cell fate, proliferation and apoptosis [16-19].…”
