Wogonin induces cell cycle arrest and erythroid differentiation in imatinib-resistant K562 cells and primary CML cells

Abstract: Wogonin, a flavonoid derived from Scutellaria baicalensis Georgi, has been demonstrated to be highly effective in treating hematologic malignancies. In this study, we investigated the anticancer effects of wogonin on K562 cells, K562 imatinib-resistant cells, and primary patient-derived CML cells. Wogonin up-regulated transcription factor GATA-1 and enhanced binding between GATA-1 and FOG-1, thereby increasing expression of erythroid-differentiation genes. Wogonin also up-regulated the expression of p21 and in… Show more

“…Cheng et al reported that magnetic nanoparticles of Fe3O4 and Wogonin overcame multidrug resistance K562/A02 cells in vitro [36]. Wogonin induced cell cycle arrest and erythroid differentiation in imatinib-resistant K562 cells and primary CML cells [37]. Enhancement of apoptosis and reduction of telomerase activity were induced by Wogonin in the HL-60 leukemia cells [38].…”

“…Many natural compounds are able to do this in combination or not with the inhibition of Bcr-Abl and finally activate the erythroid transcription factor GATA-1. Wogonin induces erythroid differentiation in K562 cells mediated by activation of GATA-1 and inhibition of Bcr-Abl [77].…”

“…Previous studies (9,10) have demonstrated the antitumor potential of wogonin, particularly its ability to induce apoptosis and its high maximum-tolerated dose. Wogonin has also been observed to be a potential anti-pancreatic cancer drug through its ability to inhibit the Mcl-1 and nuclear factor-κB (NF-κB) pathways (11,12).…”
