X chromosome painting in Microtus: Origin and evolution of the giant sex chromosomes

Abstract: Sex chromosomes in species of the genus Microtus present some characteristic features that make them a very interesting group to study sex chromosome composition and evolution. M. cabrerae and M. agrestis have enlarged sex chromosomes (known as 'giant sex chromosomes') due to the presence of large heterochromatic blocks. By chromosome microdissection, we have generated probes from the X chromosome of both species and hybridized on chromosomes from six Microtus and one Arvicola species. Our results demonstrated… Show more

“…It is unlikely that these two mechanisms of ampli¢ca-tion took place during the evolution of the sex chromosomes of M. cabrerae, since the composition of the heterochromatin of both sex chromosomes is di¡erent. In fact, di¡erences in sequence composition between the X and Y heterochromatic blocks of M. cabrerae have been revealed by FISH with MSAT-160 satellite DNA (Ferna¤ ndez et al 2001) or with SRY gene (Ferna¤ ndez et al 2002), by X chromosome painting (Marchal et al 2004) and in this study. Other ampli¢cation mechanisms, such as sister chromatid exchanges or replication slippage, could also explain the heterochromatin ampli¢cation on the sex chromosomes of Microtus.…”

“…When the DNA was amplified by this last method, we immediately reamplified it, following supplier instructions, and then marked it by nick translation with rhodamine-11-dUTP or fluorogreen-11-dUTP. When the DNA was amplified by DOP-PCR, we re-amplified and marked DNA with SpectrumOrange-dUTP (Vysis) [Marchal et al, 2004].…”

“…Chromosome painting was performed as described in Marchal et al [2004] on mitotic metaphase cells from 1 embryo obtained Teruel /Cabrero /Perfectti /Acosta / Sánchez /Camacho Cytogenet Genome Res 2009;125:286-291 288 in our laboratory carrying the B 1 (from the Mundo river population, Albacete, Spain) and 2 embryos carrying B 24 (from Torrox) chromosomes. In addition, primary spermatocytes from a male collected at Goynuk (Turkey) were submitted to chromosome painting.…”

“…In a diversity of organisms, heterochromatinization, accompanied by amplification of tandem repeats, represents an important step in the morphological differentiation of simple sex chromosome systems, especially in the ZW ones (Nanda et al, 2000;Kondo et al, 2004;Marchal et al, 2004;Peichel et al, 2004;Charlesworth et al, 2005;Ezaz et al, 2009;Kejnovský et al, 2009). Same type of studies provided evidence that rDNA cistrons can also occur on the sex chromosomes of distinct organisms (see, for example, Goodpasture and Bloom, 1975;Schmid et al, 1983;Yonenaga-Yassuda et al, 1983;Morielle and Varella-Garcia, 1988;Cioffi et al, 2010).…”
