YAP and the RhoC regulator ARHGAP18, are required to mediate flow-dependent endothelial cell alignment

Abstract: Background: Vascular endothelial cell alignment in the direction of flow is an adaptive response that protects against aortic diseases such as atherosclerosis. The RhoGTPases are known to regulate this alignment. We have shown previously that ARHGAP18 in endothelial cells is a negative regulator of RhoC and its expression is essential in flow-mediated alignment. Depletion of ARHGAP18 inhibits alignment and results in the induction of a proinflammatory phenotype. In embryogenesis, ARHGAP18 was identified as a d… Show more

“…The hub gene of the black module was ARHGAP18. ARHGAP18 (SENEX) is a negative regulator of YAP and RhoC (Coleman et al, 2020). Overexpression of RhoC destroys the actin cytoskeleton and cell junctions.…”

“…Recently, the role of ARHGAP18 has been extensively studied in the context of atherosclerosis, a chronic inflammatory disease of the arteries [ 43 ]. ARHGAP18 was shown to act as an anti-inflammatory and athero-protective gene that facilitates flow-responsive endothelial cell alignment via ARHGAP18/YAP axis [ 29 , 30 ]. Interestingly, PKN3 was identified in a module of genes associated to transendothelial migration of leukocytes leading to coronary artery disease [ 44 ].…”

“…Overexpression of ARHGAP18 leads to premature senescence of endothelial cells [ 28 ]. Recently, it was shown to facilitate athero-protective endothelial cell alignment in response to laminar shear flow [ 29 , 30 ].…”

“…Human umbilical vein endothelial cells (HUVECs) were obtained from umbilical cords by collagenase treatment [ 40 ], after donor informed consent, under Ethics approval by the Sydney Local Health District Human Ethics Committee, X16-0225. HUVECs were isolated and maintained in culture in MesoEndo growth medium (Lonza, Basel, Switzerland) in 5% CO 2 incubator.…”
