Chlorella vulgaris (C. vulgaris) powder is a novel non-grain single-cell protein with enormous potential to be a protein source. However, it is poorly studied in aquatic animals. The purpose of the present study was to explore the optimum replacement ratio of C. vulgaris powder and the influence of the substitution of soybean meal with C. vulgaris on grass carp (Ctenopharyngodon idella) in terms of growth performance, intestinal integrity and the microbial community. Five isonitrogenous and isolipidic diets were formulated by replacing 0% (SM, containing 30% soybean meal), 25% (X25), 50% (X50), 75% (X75) and 100% (X100) soybean meal with C. vulgaris. The feeding trial period lasted 8 weeks. At the end of the experimental trial, the X50 group showed higher FW, WGR and PER than the SM group (p < 0.05). The feed conversion ratio (FCR) of the X50 group was significantly lower than that of the SM group (p < 0.05). The X50 group showed the highest value of the goblet cell number, intestinal amylase and trypsin activities when compared with the SM group (p < 0.05). Replacing 50% soybean meal with C. vulgaris improved the intestinal barrier integrity, as evidenced by upregulating zo-1, zo-2 and occluding transcript (p < 0.05), and alleviated oxidative stress by an increased SOD enzymatic activity and transcript level, probably mediated through the Nrf2-keap1 signaling pathway (p < 0.05). Meanwhile, the X50 group enhanced intestinal immunity, as manifested by increased ACP and LZM activities (p < 0.05), and downregulated the tlr-4, tlr-7, tlr-8 and il-6 through the tlr pathway (p < 0.05). The functionally predicting pathways related to the nitrate respiration and nitrogen respiration were observably activated in the X50 group (p < 0.05). The X50 group improved the biological barrier, as manifested by increased Firmicutes and Rhodobacter (p < 0.05). In conclusion, dietary C. vulgaris powder could promote the growth performance of grass carp by restoring intestinal morphology, increasing digestive enzyme activities, improving antioxidant properties and immunity and optimizing the microflora structure. A C. vulgaris powder replacement of 50% soybean meal was recommended as feed for grass carp.

IntroductionThis research aimed to assess the impact of Fermented Chinese herbs (FCHM) on the intestinal barrier, immunity, and protection against Aeromonas hydrophila (A. hydrophila) infection in largemouth bass (Micropterus salmoides).MethodsFour experimental diets were formulated, including H0 (basal diet), H1, H2, and H3, which contained 1%, 3%, and 5% FCHM added to the basal diet, respectively. The fish were randomly allocated to four treatment groups, each with 3 parallel per treatment, consisting of 20 fish per replicate and were raised for 56 days.Results and discussionThe experiment revealed that: compared with the control group, adding 1% FCHM significantly improved the weight gain rate (WGR) and specific growth rate (SGR) of the juvenile largemouth bass (P< 0.05). The ingestion of FCHM substantially elevated the activities of CAT, SOD, GSH-PX, APK, ACP, and LZM, and T-AOC level in the gut region of largemouth bass, while decreasing the MDA content in intestine (P< 0.05). Supplementation with FCHM enhanced the intestinal villus height and relative mRNA expression of intestinal barrier genes ZO-1, Claudin, and Occludin in juvenile largemouth bass. After injecting A.hydrophila, all groups of largemouth bass experienced mortality, but the consumption of FCHM resulted in a decrease in cumulative mortality. After infected with A.hydrophila, the antioxidant enzymes and immune enzymes activities of all test groups were enhanced compared to those before infection, and the antioxidant enzymes and immune enzymes activities of all groups were considerably higher than the control after feeding FCHM (P< 0.05). After infected with A.hydrophila, the intestinal MDA content of largemouth bass was higher compared with that before infection in all cases (P< 0.05), but after feeding FCHM, the MDA content was lower than the control (P< 0.05). Upon consuming FCHM, the mRNA relative expressions of pro-inflammatory biomarkers IL-1β, TNF-α, IL-15 and IL-8 in largemouth bass infected with A.hydrophila were decreased in comparison to the control group. In contrast, the mRNA expression of the anti-inflammatory biomarkers TGF-β and IL-10 were significantly elevated (P< 0.05). In summary, FCHM could improve the intestinal morphology, immunity, and antioxidant capacity of juvenile largemouth bass, and enhance it against A.hydrophila, with a better effect at 1% addition.

The study aimed to evaluate the effects of fermented tea residue (FT) on growth performance, intestinal morphology, liver antioxidant capacity and Aeromonas hydrophila infection in juvenile Largemouth bass. A total of 240 fish were randomly distributed in 12 tanks with 20 fish per tank (4 treatments with 3 replications) and fed with diets FT at the rate of 0 (control), 2, 4 and 6%. The weight gain rate (WGR), specific growth rate (SGR) and intestinal villi height (VH) of juvenile largemouth bass were significantly higher than those of the control group after feeding FT (P< 0.05); meanwhile, the liver superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and catalase (CAT) activities of juvenile largemouth bass were significantly higher and the malondialdehyde (MDA) levels were significantly lower than those of the control group after feeding FT (P< 0.05). Mortality occurred in all groups of largemouth bass after the injection of A.hydrophila, but feeding FT reduced the cumulative mortality compared with the control group (P< 0.05). In juvenile largemouth bass infected with A.hydrophila, the relative mRNA expression of the intestinal anti-inflammatory factors IL-10 and TGF-α was significantly higher and that of the pro-inflammatory factors IL-1, IL-15, IL-8, and TNF-α was significantly lower (P< 0.05). In summary, it can be seen that a 2% FT addition can improve the liver antioxidant capacity of juvenile largemouth bass, enhance the resistance to A.hydrophila and increase the growth of largemouth bass.
