Yeast histone deposition protein Asf1p requires Hir proteins and PCNA for heterochromatic silencing

Abstract: Yeast CAF-I and Asf1p cooperate to form nucleosomes in vitro. In vivo, Asf1p and Hir proteins physically interact and together promote heterochromatic gene silencing in a manner requiring PCNA. This Asf1/Hir silencing pathway functionally overlaps with CAF-I activity.

“…Yeast Hir1p and Hir2p share several biological and biochemical properties with Asf1p. Like Asf1p, they are required for heterochromatin-mediated silencing of telomeres and mating loci, and are also required for formation of proper pericentromeric chromatin (but in an Asf1p-independent manner) (64,67,116,117). Mouse ES cells lacking HIRA have a larger pool of loosely bound histones than wild type cells, consistent with a role for HIRA in generation of compact, nucleosome-dense, transcriptionally silent heterochromatin (78).…”

“…In vitro, HIRA has been shown to act as a histone chaperone, preferentially depositing the histone variant histone H3.3 into nucleosomes, compared to canonical histone H3.1 (49,72,100,102,125). Consistent with their partially overlapping functions, Asf1 and Hir proteins physically interact and in yeast this interaction is necessary for telomeric silencing (32,49,116).…”

“…Significantly, at least one of these, the histone chaperone HIRA, has been shown to increase in expression or undergo some level of regulation in aging baboon skin [40,41]. Although the HIRA histone chaperone may have some functions linked to transcription activation [42][43][44][45][46][47][48], this chaperone is known to have an evolutionarily conserved role in heterochromatin formation [49][50][51][52][53][54][55][56].…”

“…In S. cerevisiae, Hir gene products (HIR1 and HIR2) are implicated in a nucleosome assembly pathway that functionally overlaps that of chromatin assembly factor-1 (CAF-I) (Sharp et al 2001). HIR1 and HIR2 have been shown to be important for normal growth and gene silencing in the absence of CAF-I (Kaufman et al 1998;Qian et al 1998).…”
