Zirconium-Based Metal–Organic Framework Nanocarrier for the Controlled Release of Ibuprofen

Abstract: Electrospray-differential mobility analysis coupled with aerosol particle mass analysis (ES-DMA/APM) was demonstrated for the development of a metal–organic framework (MOF) nanocarrier system. A successful quantification of ibuprofen loading in UiO-66-NH2 (i.e., the representative drug molecule and MOF, respectively) achieved based on the aerosol particle mass of MOF measured by ES-DMA/APM (≈55 mg of ibuprofen/g of UiO-66-NH2). The structural stability of UiO-66-NH2 versus ibuprofen release was successfully qu… Show more

“…The stability of MOF is relatively weak under a variety of conditions, including low pH which can lead to the protonation of the imidazole ligand ( Li et al, 2017 ) or rigid carboxylate ligands, various ions (MgCl 2 or NaCl-containing Tris-HCl buffer ( Zhang et al, 2014b ), PO 4 3− ( Wang et al, 2019a )) due to the anion exchange ( Velasquez-Hernandez et al, 2019 ), which leads to the break of the coordination bond and the destruction of the MOF structure, further limiting its sensing applications. Now, only a limited MOF has good water stability ( Qiu et al, 2019 ): (1) the metal carboxylate frameworks composed of high-valence metal ions (such as Cr 3+ , Fe 3+ , and Zr 4+ ) which provide high coordination number and charge density, including MIL-101(Cr), MIL-100 (Fe), UiO-66(Zr), and PCN family; (2) the metal azolate frameworks composed of soft nitrogen-donor ligands (such as imidazolates, tetrazolates, triazolates, and pyrazolates).…”

“…In addition, Pt modified electrodes have also been widely explored for the detection of hydrogen, and other gases like oxygen and ammonia in recent years. [5][6][7][8] MOFs are a new class of functional porous materials with inorganic and organic species arranged in a regular pattern [9][10][11][12] . The inorganic component in MOFs is usually a metal or metal oxide node, and the organic part is a multidentate ligand such as a benzene carboxylic acid.…”

“…To increase the biological functionality and biocompatibility functionalization of MOFs with biomolecules such as nucleobases, saccharides, peptides, and amino acids have been employed as the organic ligands to prepare bio-MOFs. Zirconium-based MOF was prepared as a nanocarrier for the controlled release of ibuprofen in an acidic phosphate buffer solution [ 226 ]. Horcajada and collaborators reported the use of a series of non-toxic porous iron (III)-based nanoMOFs [ 227 ], i.e., MIL-53, MIL-88A, MIL-88Bt, MIL-89, MIL-100 and MIL-101-NH 2 , as carriers of antineoplastic and retroviral drugs, including busulfan, azidothymidine triphosphate, doxorubicin or cidofovir, ibuprofen, etc.…”
