Zoospore chemotaxis in Australian isolates of Phytophthora species

Abstract: Zoospores of Australian isolates of Phytophthora drechsleri, P. cryptogea, P. cinnamomi, P. nicotianae var. parasitica, and P. citricola were examined for their chemotactic responses to asparagine, glutamine, aspartate, glutamate, and structurally related compounds. Structural requirements for attraction include the alpha-amino-acid group with a short carbon chain terminating in an amide group. The one American isolate tested gave a different result and possible reasons for this are discussed. The pH of the en… Show more

“…For example, taxis occurs towards root difFusates and mixtures of nutrients, and, depending on the fungus, towards individual sugars and amino aeids (e.g. Royle & Hiekman, 1964i;Ho & Hickman, 1967;Chang-Ho & Hiekman, 1970), ethanol (Allen & Newhook, 1973;Halsall, 1976) and other volatile compounds (Cameron & Carlile, 1978). It occurs also in response to eleetrieal or ionic fields (Troutman & Wills, 1964;Khew & Zentmyer, 1974;Miller, Shand & Gow, 1988).…”

“…Simulated root models, complex mixtures such as V8 broth, and some pure compounds and defined mixtures have been used to demonstrate various parts of the sequence in vitro (Dukes & Apple, 1961;Royle & Hickman, 1964a, b ;Zentmyer, 1966;Bimpong & Clerk, 1970;Ho & Hickman, 1967;Khew & Zentmyer, 1973;Halsall, 1976). In general no pure compound approached the activity of root exudate and most were only effective at high concentrations.…”

“…Zoospores are chemotactically attracted to root tips (Halsall, 1976) and to points where lateral roots are about to emerge: on root tips they attach and encyst mainly in the zone of elongation just behind the root cap. Germ tubes from the encysted spores then penetrate the root tissue.…”

“…In water, sporangia from oospores are small and release 8-16 zoospores (Duncan, 1975); larger sporangia (on average 50 × 30 µm (Bain and Demaree, 1945)) are produced on infected roots and can release 40-50 zoospores, perhaps more. Zoospores are chemotactically attracted to root tips (Halsall, 1976) and to points where lateral roots are about to emerge: on root tips they attach and encyst mainly in the zone of elongation just behind the root cap. Germ tubes from the encysted spores then penetrate the root tissue.…”
