Zscan4 Inhibits Maintenance DNA Methylation to Facilitate Telomere Elongation in Mouse Embryonic Stem Cells

Abstract: SUMMARY Proper telomere length is essential for embryonic stem cell (ESC) self-renewal and pluripotency. Mouse ESCs (mESCs) sporadically convert to a transient totipotent state similar to that of two-cell (2C) embryos to recover shortened telomeres. Zscan4, which exhibits a burst of expression in 2C-like mESCs, is required for telomere extension in these cells. However, the mechanism by which Zscan4 extends telomeres remains elusive. Here, we show that Zscan4 facilitates telomere elongation by inducing global … Show more

“…Elevated DUX level in turn increases bipotent 2C-like cells in ESC culture through global transcriptional activation of 2C-specific genes, including genome caretaker genes, Zscan4 and Tcstv1/3 8,38 . In addition to their role in telomere elongation through activation of telomere sister chromatid exchange 7,8 , Zscan4 genes can also promote DNA repair by facilitating heterochromatin decondensation and DNA demethylation 6,15,39 . Therefore, ATR-mediated activation of Zscan4 genes likely contributes to ESCs genomic integrity in response to RS.…”

“…Southern blot, dot blot and IF analyses of DNA methylation were performed as described previously (Dan et al, 2017). For bisulfite sequencing analysis, bisulfite conversion was performed using EZ DNA Methylation Kit (Zymo), PCR products cloned using NEB PCR cloning kit, and individual clones sequenced.…”

“…To circumvent this, we took advantage of a spontaneously occurring rare subpopulation of primed mouse embryonic stem cells (ESCs) that express transcripts usually restricted to ZGA, including the MERVL endogenous retrovirus and Zscan4 cluster (Macfarlan et al 2012;Zalzman et al 2010;Ishiuchi et al 2015;Eckersley-Maslin et al 2016). These "2C-like" ESCs also share several epigenetic features with the two-cell embryo, including global DNA hypomethylation (Eckersley-Maslin et al 2016;Dan et al 2017), decondensed chromatin (Eckersley-Maslin et al 2016;Ishiuchi et al 2015;Akiyama et al 2015) and increased histone mobility (Boskovic et al 2014).…”
