Zymographic Analysis and Characterization of MMP-2 and -9 Forms in Human Sound Dentin

Abstract: The role and function of dentin matrix metalloproteinases (MMPs) are not well-understood, but they may play a key role in dentinal caries and the degradation of resin-bonded dentin matrices. To test the null hypothesis that MMP-9 is not found in dentin matrix, we used gelatin zymography to extract and isolate all molecular forms of gelatinolytic MMPs in demineralized mature sound dentin powder obtained from extracted human molars, characterizing and identifying the enzymes by Western blotting. Gelatinolytic MM… Show more

“…Following electrophoresis, the gel was divided into equal parts, and the individual strips were developed in different developing buffers containing 5 lg/ml CSE, 10 lg/ml CSE, or 1 mM EDTA as positive control, respectively (Albini et al 2001;Mazzoni et al 2007). …”

“…[1][2][3][4][5][6][7] A correlation between the presence of active endogenous matrix metalloproteinases (MMPs) in dentin matrices and premature degradation of hybrid layers has been demonstrated. 8,9 Different forms of MMPs have been identified in dentin matrices by zymographic analyses, with MMP-2 and MMP-9 being the most prevalent. 8,10,11 When exposed to an acidic environment, such as the one created by the application of acidic adhesive resins, these enzymes, initially secreted as pro-enzymes, become active proteinases.…”

“…More recently, MMPs were identified as being responsible for that activity (Tjäderhane et al 1998), and to date, the presence of gelatinases MMP-2 and -9 ( Fig. 1), collagenase MMP-8, stromelysin MMP-3, and MMP-20 have been reported (Martin-De Las Heras et al 2000;Sulkala et al 2002;Mazzoni et al 2007;Sulkala et al 2007;Boukpessi et al 2008;Mazzoni et al 2009;Santos et al 2009;Boushell et al 2011;.…”
