Zymography Methods to Simultaneously Analyze Superoxide Dismutase and Catalase Activities: Novel Application for Yeast Species Identification

Abstract: We provide an optimized protocol for a double staining technique to analyze superoxide dismutase enzymatic isoforms Cu-Zn SOD (Sod1) and Mn-SOD (Sod2) and catalase in the same polyacrylamide gel. The use of NaCN, which specifically inhibits yeast Sod1 isoform, allows the analysis of Sod2 isoform while the use of HO allows the analysis of catalase. The identification of a different zymography profiling of SOD and catalase isoforms in different yeast species allowed us to propose this technique as a novel yeast … Show more

“…Active enzymes such as SOD are known to participate in the reduction of ROS contributing to the reduction of inflammation. SOD activity can be rapidly visualized using zymography [ 13 , 14 ]. SOD activity was measured in Thykamine extracts with white bands observed on the SDS-PAGE gel obtained by a decreased in O 2 − formed by the reaction between nitro blue and riboflavin ( Figure 2 D).…”

“…SOD zymography was performed in non-denaturating conditions on a polyacrylamide gel (acrylami-de/bis-acrylamide 30%/10%) [13,14]. Thykamine extracts were added to obtain a final concentration of 0.2 mg proteins.…”

“…Equal amounts of protein were loaded onto a 10% non-denaturing polyacrylamide gel and were run under non-denaturing conditions. In-gel catalase activity was visualized as previously described ( Gamero-Sandemetrio et al., 2017 ). This analysis was carried out twice using separate biological replicates each time.…”
