α 2-Macroglobulin and fibrinogen modulate inflammatory edema in man

Abstract: Animal experiments suggest that the response of acute-phase proteins (APPs) modulates the inflammatory reaction following tissue injury. To study this in man we investigated the relation between a number of APPs, including fibrinogen and alpha 2-macroglobulin, and the inflammatory edema induced by a primary immunization against cholera, typhoid, and yellow fever. Vaccination induces a significant APP response; however, only alpha 2-macroglobulin and fibrinogen were of importance to the amount of edema, measure… Show more

“…They also showed that values during the steady state in patients with rheumatoid arthritis were lower than in controls (49), again suggesting that a 2 M being removed from the circulation in these inflammatory processes seems similar. From a different point of view, one group has shown that a 2 M levels correlate inversely with the amount of tissue swelling after immunization for infectious diseases (50). The higher the levels of a 2 M, the less local edema, in effect suggesting that the antiproteolytic effect of a 2 M reduces local edema.…”

“…There are many reports describing the effects of clusterin, haptoglobin and α 2 M on the immune system (59,(176)(177)(178)(179)(180). Some of these, such as the ability of α 2 M to enhance antigen presentation (91) and haptoglobin-facilitated clearance of hemoglobin (180), clearly fall outside of the scope of this review and as such, will not be discussed here.…”
