α-Glucan, water dikinase (GWD): A plastidic enzyme with redox-regulated and coordinated catalytic activity and binding affinity

Abstract: The recently discovered potato tuber (Solanum tuberosum) ␣-glucan, water dikinase (GWD) (formerly known as R1) catalyzes the phosphorylation of starch by a dikinase-type reaction mechanism in which the ␤-phosphate of ATP is transferred to either the C-6 or the C-3 position of the glucosyl residue of starch. In the present study, we found that the GWD enzyme is inactive in the oxidized form, which is accompanied by the formation of a specific intramolecular disulfide bond as determined by disulfide-linked pepti… Show more

“…First, enzymes shown to be regulated by Trx reside in the organelle, namely, ADP-glucose pyrophosphorylase (18), NADP-glucose 6-phosphate dehydrogenase (19,20), and ␣-glucan, water dikinase (21). Second, analyses of the constituent proteins (proteome) of amyloplasts have revealed the presence of a number of other enzymes that, in chloroplasts, are linked to Trx (refs.…”

“…The diurnal accumulation of starch thus seems to incorporate three regulatory elements: protein phosphorylation, redox, and pH. Redox is known to regulate other enzymes functional in starch metabolism, e.g., thioredoxin controls the activity of ADP-glucose pyrophosphorylase (18,19), the activity and binding of ␣-glucan, water dikinase (SEX1) to starch granules (20), and possibly the activity of ␤-amylase (21). However, to our knowledge, this is the first case in which redox has been linked to an enzyme involved with protein phosphorylation for the regulation of starch metabolism.…”

“…15) Other GWDs also play an essential role in plastidial starch turn over in leaves as well as in potato tubers. 14,31,32) The debranching enzyme barley limit dextrinase. Barley limit dextrinase (LD), the key enzyme in α limit dextrin debranching and with high activity also on the linear polysaccharide pullulan, 18 21) has a major role in starch mobilization during seed germination and is possibly also involved in starch biosynthesis by trimming of intermediate branched α glucan structures.…”
