α‐Tomatine, the major saponin in tomato, induces programmed cell death mediated by reactive oxygen species in the fungal pathogen Fusarium oxysporum

Abstract: The tomato saponin a-tomatine has been proposed to kill sensitive cells by binding to cell membranes followed by leakage of cell components. However, details of the modes of action of the compound on fungal cells are poorly understood. In the present study, mechanisms involved in a-tomatine-induced cell death of fungi were examined using a filamentous pathogenic fungus Fusarium oxysporum. a-Tomatine-induced cell death of F. oxysporum (TICDF) occurred only under aerobic conditions and was blocked by the mitocho… Show more

“…Recently, Ito et al [1] found that the antibiotic effect of tomatine against the fungal pathogen Fusarium oxysporum involves activation of phosphotyrosine kinase and G-protein signaling pathways leading to Ca 2+ elevation and accumulation of reactive-oxygen species (ROS). In related studies, Simons et al [2] found that the mode of action of atomatine towards yeast cells involving cell membrane permeilization is distinct from that of the aglycone tomatidine that lacks the tetrasaccharide side chain.…”

“…Apoptosis has been observed in a number of filamentous fungi, such as Aspergillus nidulans, Aspergillus fumigatus, Neurospora crassa, and Fusarium oxysporum (12)(13)(14)(15). Various physiological stresses, ionizing radiation, and hyperthermia have been reported to trigger apoptosis in fungi and cancer cells (14)(15)(16).…”

“…It is interesting that in F. oxysporum f. sp. lycopersici, the observed sensitivity to ␣-tomatine is a result of ␣-tomatine-induced programmed cell death in the fungus rather than an effect on membranes (23). Furthermore, the ␣-tomatine aglycone tomatidine inhibits sterol biosynthesis in Saccharomyces cerevisiae (50), indicating that the biological properties of saponins and their degradation products are complex.…”
