α1,3 Glucans Are Dispensable in Aspergillus fumigatus

Abstract: A triple ␣1,3 glucan synthase mutant of Aspergillus fumigatus obtained by successive deletions of the three ␣1,3 glucan synthase genes (AGS1, AGS2, and AGS3) has a cell wall devoid of ␣1,3 glucans. The lack of ␣1,3 glucans affects neither conidial germination nor mycelial vegetative growth and is compensated by an increase in ␤1,3 glucan and/or chitin content.

“…Even though the cell wall of the triple ags mutant is devoid of a (1,3) glucan, fungal growth and conidiation is not affected in vitro [25]. However, this mutant was avirulent in neutropenic mice due to a block in conidial germination [20 ].…”

“…Fission yeast contains four additional Ags1 homologues (Mok11 to Mok14), which are detected only during sporulation (81). Ags1 orthologues are not found in budding yeasts but are widely extended in filamentous, dimorphic, and pathogenic fungi (82,83).…”

“…Among the Aspergillus species, the functions of α-1,3-glucan synthase genes have been studied in A. fumigatus and A. niger [11][14]. Aspergillus fumigatus contains three AGS genes, ags 1 to ags 3.…”

“…The expression of agsA (an orthologue of A. fumigatus ags3 ) and agsE (an orthologue of A. fumigatus ags1 and A. nidulans agsB ) was induced in the presence of cell wall stress–inducing compounds such as calcofluor white (CFW), sodium dodecyl sulfate, and caspofungin [13]. More recently, a triple-mutant strain of A. fumigatus lacking the three α-1,3-glucan synthase genes ( ags1 , ags2 , and ags3 ) was generated, and growth of the triple mutant in plate culture was similar to that of the parental strain [14]. The triple mutant showed slightly decreased conidiogenesis, as did the single ags1 and ags2 mutants [11], [14], and the lack of cell wall α-1,3-glucan led to an increase in β-1,3-glucan and chitin levels in mycelia of the triple mutant [14].…”
