β-Adrenergic Receptor Inhibitor and Oncolytic Herpesvirus Combination Therapy Shows Enhanced Antitumoral and Antiangiogenic Effects on Colorectal Cancer

Abstract: Oncolytic viruses (OVs) are considered a promising therapeutic alternative for cancer. However, despite the development of novel OVs with improved efficacy and tumor selectivity, their limited efficacy as monotherapeutic agents remains a significant challenge. This study extended our previously observed combination effects of propranolol, a nonselective β-blocker, and the T1012G oncolytic virus into colorectal cancer models. A cell viability assay showed that cotreatment could induce synergistic killing effect… Show more

“…Also, propranolol and T1012 combined therapy promoted vascular endothelial growth factor secretion inhibition. However, this combined treatment does not promote viral replication compared to that of cetuximab in an animal study (Hu et al, 2021a).…”

“…OVs also stimulate the production of CXCL9 and CXCL10, providing critical signals for T cell trafficking. Furthermore, OVs can induce degradation of the tumor stroma, breaking down physical barriers to T cell infiltration [ 32 ]. However, using OVs alone may not sustain long-term immune responses and may lead to immune resistance.…”

“…Besides direct lysis of cancer cells, OVs attack tumor vasculature and eventually lead to thrombosis and vascular atrophy, leading to ischemia and hypoxia in cancer cells and reducing subsequent viral spread within tumor tissue. 116 The strategy of lyssavirus attack on the tumor vascular system is to target vascular endothelial cells within the tumor while selectively enhancing tumor vascular permeability. 117 An experimental study showed that oncolytic adenovirus can infect, replicate, and expand in tumor-associated endothelial cells, causing inhibition of tumor angiogenesis.…”
