β‐caryophyllene oxide inhibits constitutive and inducible STAT3 signaling pathway through induction of the SHP‐1 protein tyrosine phosphatase

Abstract: Constitutive activation of STAT3 is frequently observed and closely linked with proliferation, survival, invasion, metastasis and angiogenesis in tumor cells. “…β ‐Caryophyllene still inhibits the PI3K/AKT/mTOR/S6K1 pathway, suppressing the expression of phospho‐AKT (Ser473), in a concentration‐dependent manner, in PC‐3 and MCF‐7 cancer cells, and was capable of blocking the IL‐6‐induced activation of STAT‐3 and the upstream elements of the STAT3 pathway (Src, JAK1, and JAK2) in time‐ and dose‐dependent manners, by the expression of tyrosine phosphatase SHP‐1. This indicates that BCP could interfere with TORC2 (the mTOR‐rictor complex that phosphorylates AKT at Ser473) .
