β-Catenin regulates airway smooth muscle contraction

Abstract: beta-Catenin is an 88-kDa member of the armadillo family of proteins that is associated with the cadherin-catenin complex in the plasma membrane. This complex interacts dynamically with the actin cytoskeleton to stabilize adherens junctions, which play a central role in force transmission by smooth muscle cells. Therefore, in the present study, we hypothesized a role for beta-catenin in the regulation of smooth muscle force production. beta-Catenin colocalized with smooth muscle alpha-actin (sm-alpha-actin) an… Show more

“…Besides canonical WNT ligand, TGF-β also stabilizes β-catenin where it participates in TGF-β-specific cellular responses [38]. Our group has previously identified important physiological and functional roles for β-catenin in airway smooth muscle cells [36], [39][41]. Here, we describe a novel role for β-catenin in WNT-5A induction.…”

“…Indeed, we describe that treatment of several melanoma cell lines and HCT116 inhibit both, β-catenin expression and transcriptional activity in a dose-dependent fashion. More recent publications described also that PKF115–584 treatment resulted in a dose-dependent reduction of total beta-catenin protein levels in some acute myeloid leukemia cell lines [42], smooth muscle cells [43] and mouse embryonic fibroblasts [44]. All of the available data indicate that PKF115–584 may be not a selective inhibitor of the TCF/beta-catenin complex, but can inhibit also total beta-catenin protein levels.…”
