“…While β-lactams, i.e., azetidin-2-ones, are a rich source of antibiotics, [2] their structural isomer, azetidin-3-ones, with the carbonyl group one-carbon removed from the nitrogen atom, have not been found in nature but could serve as versatile substrates for the synthesis of functionalized azetidines. “…It is widely accepted that the high reactivity of b-lactam antibiotics results from the lack of amide resonance in the 2-azetidinone ring [1][2][3]. In penams (1), penems (2, X ¼ S) and carbapenems (2, X ¼ CH 2 ), the p electrons of the N-C]O function cannot be delocalized over the three atoms owing to the structural constraints which prevent the amide substituents to lie in the same plane.…”

“…In the field of antibiotics, [2] a lot of works has been devoted to the search of more strained, fused bicyclic b- [10,11] and g- [12,13] lactam systems.…”

“…A bacterial cell wall obtains mechanical stability by cross‐linking carbohydrate co‐polymer chains built from N ‐acetylglucosamine and muraminic acid with peptide bridges to form murein. β‐Lactam antibiotics inhibit this process by inhibition of the transpeptidase enzyme carrying out the cross‐linking, while vancomycin binds specifically to the interpeptide sequence . The compounds to be developed here should share the same biological activity and mechanism as vancomycin, being selected to bind to the specific amino acid sequence of the interpeptide linkage.…”
