γδ T cell homeostasis is established in competition with αβ T cells and NK cells

Abstract: γδ T cells are a diverse population of lymphocytes that play an important role in immune regulation. The size of the γδ T cell pool is tightly regulated, comprising only 1-10% of total lymphoid T cells in mice and humans. We examined the homeostatic regulation of γδ T cells using a model of lymphopenia-induced homeostatic expansion. We found that IL-15 and, to a lesser extent, IL-7 play an important role in lymphoid γδ T cell homeostasis. Moreover, γδ T cell homeostatic expansion was limited not only by γδ T c… Show more

“…We adapted this system to test the effect of Treg on gd-cell expansion in Rag2-deficient animals; we added P. berghei ANKA infection as an extra stimulus (1 day after the injection of lymphocytes) due to the relatively poor homeostatic expansion of gd cells (data not shown) [32]. In this adapted model, Teff and Treg engrafted and expanded comparably after 4 days ( Fig.…”

“…Since the lack of B cells influenced the number of splenic NK cells, which are dependent on IL-15 for their survival [21][22][23], we tested whether B cell deficiency had an impact on T cell populations, which similarly depend on IL-15 [5,24,26]. In addition to NK cells, the numbers of TCRcd were significantly decreased in lMT -/-as compared to B6 mice (Fig.…”

“…Similar to NK cells, memory CD8 T cells [5,24], TCRab NKT cells [25] and TCRcd cells [26] depend on IL-15 for their peripheral maintenance. However, while TCRcd and NK cells were reduced, the proportion of CD8 + memory T cells (of naive CD8 + T cells) is normal in B cell-deficient mice.…”

“…This expansion is driven by IL-7 and IL-15 of which the latter also regulates the homeostasis of NK cells [47,48]. CD8 1 memory effector T cells are known to restrict IL-15-dependent homeostasis of gd-T cells [49,50]. Furthermore, NK cells and CD8 1 T cells compete for IL-15 in lymphopenic mice [51].…”
