Toxicobiological effect of mycotoxins association of the Penicillium and Fusarium fungus (T-2 toxin at a concentration of 0.1 mg/kg, fumonisin B1 ‒ 0.5 mg/kg, vomitoxin (DON) ‒ 0.1 mg/kg, penicillic acid ‒ 1 mg/kg) was accompanied by the development of a complex pathological process in weaned piglets. In this regard, the detoxification and sorption capacity of the complex feed additive "Harufix+" based on mannanoligosaccharides was studied. The additive effect on the resorptive activity of mineral and vitamin nutrients of feed under the normal feeding conditions and in case of contamination with mycotoxins has been studied. The use of enterosorbent offset the toxic effects of micromycete metabolites, which contributed to the growth of piglets. Thus, weight growth rate increase of the piglets in group 1 (i.e., those whose diet included the additive, unlike the diet of the animals in control group) constituted 16 %, while their average weight growth rate was high and constituted 1.96 kg per day. In addition, during the study of calcium, phosphorus, magnesium, ferum, zinc, copper and manganese in the piglets blood was not found excretion of these elements with a sorbent, moreover, noted the normalization their blood level. The study content of vitamins A and E, the same as with mineral nutrients, has not been established decrease during treatment with study the pharmaceutical. The obtained results testify the active absorption in the gastrointestinal tract of the vitamin components within the fodder combined with fodder additive “Harufix+” and high biological accessibility of its transport forms. The efficiency of the additive can be explained by its composition, namely the complex of mineral and organic components that are formed by modification of the organic cations of the mineral surface. The use of nanomaterials in poultry farming has become increasingly popular as feed additives capable of increasing the productivity of the herd. Nanocrystalline cerium dioxide belongs to nanomaterials with a broad spectrum of activity. Owing to their oxygen non-stoichiometry and low toxicity, cerium dioxide nanoparticles are a very promising object for agriculture. The paper presents the results of studying the meat productivity of quails of the Pharaoh breed when drinking nanocrystalline cerium dioxide in the composition of the feed additive “Nanocerium”. The study was carried out on quails during the rearing period from 14 to 49 days. For the experiment, two groups (control and experimental) of 24 heads each were formed from one day old poultry. The quails were kept in a vivarium, in cage batteries, subject to the accepted microclimate conditions. Poultry of both groups received compound feed, developed taking into account age and physiological characteristics. With drinking water, the of the experimental group additionally received nanocrystalline cerium dioxide as part of the feed additive “Nanocerium” at a dose of 8,6 mg per liter of water for 35 days. The quail population was counted and weighed weekly, starting from the day-old age. The following indicators were determined: safety, dynamics of live weight, average daily gain in live weight. According to the results of the control slaughter and anatomical cutting, the weight of the carcass and the weight of the edible part were determined. Slaughter qualities and morphological composition of quail carcasses were determined by anatomical cutting with the measurement of the following parameters: pre-slaughter weight, half-battered carcass weight, carcass patrana, slaughter yield, weight of edible parts. Drinking the feed additive “Nanocerium” by young quails increased their safety by 4.17%, live weight and absolute average daily gain by 20.3 g and 0.48 g (P <0.05), respectively. The addition of NDC quails to drinking water contributed to an increase in their pre-slaughter weight by 19.3 g, half-gutted carcass by 18.0 g, and gutted by 17.5 g in comparison with the control group. The quail carcasses of the experimental group had a higher yield of edible parts in comparison with the poultry that consumed pure water. The results of optimization of the Staphylococcus spp. identification protocol by polymerase chain reaction with agarose gel detection and approbation of the protocol with wild strains selected from dogs are presented. Determination of the parameters of specificity and sensitivity of the method was performed on museum strains of cocci S. epidermidis ATCC 14990, S. aureus ATCC 25923, S. aureus subsp. aureus UKM B-918, S. pneumoniae ATCC 49619 and E. faecalis ATCC 194433. DNA extraction was performed using the IndiSpin Pathogen Kit. The ready PCR mix NEB OneTaq® 2X Master Mix with Standard Buffer was used to prepare the reaction mixture. Primers targeted to the tuf gene region using an amplification product of 370 bp were used for the study. The reaction results were recorded in a 2% agronomic gel with the addition of ethidium bromide at a concentration of 0.5%. The optimal annealing temperature was determined by the temperature gradient method. In a study of the specificity of the method, three museum strains of staphylococci were identified as positive, while strains of other cocci did not give reaction products. The sensitivity study of the method was to detect the amplification product in seven dilutions of bacterial suspension that meet McFarland turbidity standards, the lowest concentration was further diluted 10, 100 and 1,000 times. The last dilution, which showed the presence of the amplification product corresponds to 2×106 CFU in 200 μl of saline used for DNA isolation. PCR protocol was tested on wild staphylococcal strains. Ear and nasal swabs of dogs, as well as washes from the transfer cage were selected for the study. The primary inoculation of the material was carried out on mannitol salt agar, on this medium only the growth of halophilic microorganisms is possible. Growth was found on 17 Petri dishes. The PCR washings of these cups indicated the presence of staphylococci in the test materials. The results of in-laboratory PCR testing indicate that the primer we used gives high indicators of specificity and sensitivity. Our tested technique can be used to confirm the presence of Staphylococcus spp. bacteria in the primary culture of smears taken from dogs. Conducted research pertains to technologies of animal products processing and the development of biotechnological methods for stabilizing microbial and enzymatic biological leaven for meat.Materials and Methods. The studies were conducted in the conditions of the Research Institute of Food and Animal Processing Technologies and Laboratory of Microbiological Research Methods, Department of Microbiology of Bila Tserkva National Agrarian University. Moisture retaining capacity and bacterial contamination of beef and pork were determined.Results. The effect of processing raw meat with lactic acid product – yogurt on the moisture retaining capacity was experimentally established. With the highest titratable acidity of yoghurt of 100-110 °T, the moisture retaining capacity of pork and beef was inferior to control on 2.13 and 1.29 % and amounted to 51.88 and 62.73 %. Under a titratable acidity of lactic acid beverage of 100-110 °T with total number of lactic acid bacteria on the surface of raw meat, it was 8.1×108 and 8.5×108, which prevailed samples with acidity level of 60-70 °T on 24.5 and 24.3 %.Conclusions. The results of studies have revealed the effect of processing raw meat by yogurt with different titratable acidity on moisture retaining capacity and bacterial contamination.
Intensive poultry farming technologies are closely linked to a variety of environmental, technological, feed and biological stressors, which tend to negatively affect their productivity and immune defenses. In the pathogenesis of such detrimental effects, the leading role belongs to the overproduction of oxygen free radicals - oxidative stress. The latter activates a number of transcription factors, including Nrf-2 and Nf-kB, which modulate the antioxidant defense network and participate in the organization of infection control. NDC is able to reduce the activation of Nf-kB and thus maintain antioxidant balance, but the response of the immune system to this factor is insufficiently studied. The aim of our work is to investigate the effect of NDC on the antigen of non-specific immunity of quails by adding it to drinking water. The object of study were the quail of the Pharaoh breed, experimental and control groups formed at the age of one day on the principle of analogues. The birds were kept in cages with free access to food and water. Quails of the experimental group in drinking water was added to the feed additive Nanocerium at a dose of 8.6 mg per liter of drinking water. This additive is an aqueous dispersion of NDC with an average nanoparticle size of 2-7 nm. The average weight of quails at the end of the experiment (56 days) in the experimental group was 20.2 g greater than in the control. Morphometric studies of the thymus, bursa and spleen showed no effect of NDC on the central and peripheral immune systems. Morphological parameters of the blood were within the physiological norm, but in the experimental group the number of erythrocytes and hemoglobin content were higher. The leukocyte count showed an increase in the leukocyte count (according to Garkavi LH) in the experimental group. Humoral performance was identical in both groups. Studies of cellular defense indicate no effect on the phagocytic function of peripheralblood cells. Functional and metabolic activity under the influence of NDC in the spontaneous test probably did not differ and increased in the stimulated. Therefore, quails that received nanocerium feed additive with water had a higher immunoresistance.
