Regular monitoring on experimental animal management found the fluctuation of ART outcome, which showed a necessity to explore whether superovulation treatment is responsible for such unexpected outcome. This study was subsequently conducted to examine whether superovulation treatment can preserve ultrastructural integrity and developmental competence of oocytes following oocyte activation and embryo culture. A randomized study using mouse model was designed and in vitro development (experiment 1), ultrastructural morphology (experiment 2) and functional integrity of the oocytes (experiment 3) retrieved after PMSG/hCG injection (superovulation group) or not (natural ovulation; control group) were evaluated. In experiment 1, more oocytes were retrieved following superovulation than following natural ovulation, but natural ovulation yielded higher (p < 0.0563) maturation rate than superovulation. The capacity of mature oocytes to form pronucleus and to develop into blastocysts in vitro was similar. In experiment 2, a notable (p < 0.0186) increase in mitochondrial deformity, characterized by the formation of vacuolated mitochondria, was detected in the superovulation group. Multivesicular body formation was also increased, whereas early endosome formation was significantly decreased. No obvious changes in other microorganelles, however, were detected, which included the formation and distribution of mitochondria, cortical granules, microvilli, and smooth and rough endoplasmic reticulum. In experiment 3, significant decreases in mitochondrial activity, ATP production and dextran uptake were detected in the superovulation group. In conclusion, superovulation treatment may change both maturational status and functional and ultrastuctural integrity of oocytes. Superovulation effect on preimplantation development can be discussed.
Purpose: Knowledge of nutritional support and attitudes of medical staff both affect patient care. We identified attitudes regarding nutritional support among medical staff as well as changes in attitudes when a nutritional support team is present. Methods: Data were collected from 172 medical staff members who served at a university hospital located in Gyeongju by self-administered questionnaire from June 12∼August 30, 2013. The questionnaire inquired about importance of nutritional support, self-confidence about nutritional support, consideration of nutritional support for patient care, consideration of nutritional support when a nutritional support team is present, and nutritional knowledge training during the past year. Results: A total of 169 subjects (98.3%) thought that nutritional support is important for patient care. Only 19 subjects (11.0%) were highly self-confident about nutritional support. In total, 147 subjects (85.5%) considered nutritional support for some or all patients, whereas 169 (98.3%) considered nutritional support for patient care when a nutritional support team is present. Thirty-eight subjects (22.1%) received nutritional knowledge training during the past year. High self-confidence for nutritional support was related to nutritional knowledge training and service part. Positive changes regarding nutritional support when a nutritional support team is present were associated with nutritional knowledge training and high self-confidence for nutritional support. Conclusion: No differences in nutritional support attitudes for patient care were observed among the medical staff. Nutritional knowledge training and service part affected self-confidence of nutritional support and induced positive changes in attitudes for nutritional support.
The aim of this study was to investigate the effect of high temperature on the viability of probiotic organisms (Bacillus subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae) mixed with animal feed under controlled conditions by simulating a farm feed bin in the summer. Following inoculation of probiotics into the feed, the pH and probiotic viability were monitored during an 8-day incubation at room temperature. Sterile and non-sterile feeds displayed different patterns of pH changes, with increased pH in non-sterile feed at 2 days, but a pattern of decreasing pH at 4 days. The viabilities of S. cerevisiae and B. subtilis after mono/co-inoculation were maintained without substantial changes during the incubation, whereas L. plantarum viability tended to decline. In both non-sterile and sterile feeds, the probiotics were maintained or grew without any antagonistic effects. Probiotic viability was also tested upon a shift to high temperature (60℃). There was no distinct change in pH between sterile and non-sterile feeds after the temperature shift. L. plantarum and S. cerevisiae could not survive at the high temperature, whereas B. subtilis displayed normal growth, and it inhibited the growth of contaminant microbes. Fungal growth was not observed in non-sterile feed 2 days after supplementation with B. subtilis. Therefore, heat resistant B. subtilis could be safely used in feed bins to inhibit microbial contamination, even at high temperatures. The prevention of elevated temperature in feed bins is necessary for the utilization of L. plantarum and S. cerevisiae during the summer season.
