This article describes the clinical manifestation of natural Vibrio anguillarum infection in rainbow trout (Oncorhynchus mykiss) during an outbreak on a fish farm. (i) Using an integrated approach, we characterized the pathogenesis of vibriosis from the morphological, hematological, and biochemical points of view. The molecular mechanisms associated with the host immune response were investigated using mass spectrometric analysis of trout plasma proteins. (ii) According to the severity of infection (the extent of tissue damage, the level of expression of pro-inflammatory genes, and changes in the leukocyte profile) three fish populations were identified among infected trout: fish with severe lesions (SL), fish with the moderate infectious process (IP) and asymptomatic fish (AS). (iii) Lymphopenia, granulocytosis, and splenomegaly were strong trends during the progression of infection and informative indicators of severe manifestation of disease, associated with hemorrhagic shock, metabolic acidosis, and massive tissue damage. (iv) As expected, pro-inflammatory interleukins, complement components, acute phase proteins, and antimicrobial peptides were implicated in the acute pathogenesis. Systemic coagulopathy was accompanied by increased antithrombotic reactions. (v) Reconstruction of metabolic pathways also revealed a high energy requirement for the immune response in severely affected fish. (vi) An unexpected result was a small difference between fish with moderate symptoms and fish with no or minor external signs of pathology (putatively resistant to infection). Increased production of antiproteases and enhanced blood coagulation cascade were observed in healthier fish, which may underlie the mechanisms of a controlled, non-self-damaging immune response to infection. (vii) Depending on the progression of the disease and the presence of the pathogen, a stepwise or linear change in the abundance of some plasma proteins was revealed. These proteins could be proposed as molecular markers for diagnosing the health and immune status of trout when cultured in fish farms.
The subject of this research is the process of converting former industrial sites to residential facilities. The object is the housing stock of Moscow. The author explores the theoretical aspects of reproduction of housing stock, giving special attention to conversion of the category of industrial buildings to residential category, which expands the quantitative characteristics of the housing stock. Analysis is conducted on the statistical data of conversion of industrial sites to residential facilities, as well as the available examples of redevelopment of the industrial sites. The conclusion is made that the housing stock is an interlink between many life spheres and the development of its characteristics is required on a permanent basis, which in essence is the reproduction of housing stock through reconstruction, modernization or total building renovation. The implementation of such method as conversion of former industrial sited into residential facilities would contribute to the expansion of the real estate market and increase in the number of residential units, minimizing many issues in the housing stock. The author proves the feasibility of solving issues in the housing stock of the city through conversion of former industrial sites to housing facilities. The novelty of this research lies in the fact that many authors view such method as the solution of disused former industrial buildings, rather than from the perspective of minimization of issues in the housing stock.
D-lactic acid stereoisomer (D-lactate) is produced by the intestinal microflora and can enter the bloodstream and cause in some cases a condition of acute D-lactic acidemia known as short gut syndrome. The level of D-lactate in blood and in the contents of the intestine is considered as a promising marker of the development of inflammation associated with microflora disorders, as well as with the development of a bacterial infection, while the mechanism of its entry into the blood of vertebrates from the intestine has not been studied in detail.The aim of the study. To investigate the relationship between the level of D-lactate in blood and in the intestine, taking into account the permeability of the intestinal epithelium.Materials and methods. As a model object of the study, we used juvenile rainbow trout O. mykiss. For 54 days, they were high-carbohydrate or high-protein fed. Since different types of bacteria prefer different substrates, it was expected that at the end of the experiment, the composition of the intestinal microflora would be significantly different in fish fed with different diets. The content of D-lactate in blood plasma in vitro was assessed by the Larsen method with modifications; intestinal permeability was assessed by the intensity of fluorescence of the FITС-Dextran stain in the blood of fish. The analysis of the metagenome of samples of the contents and epithelium of the fore and hind intestine was carried out. The hematological profile was partially characterized using blood smears taken immediately after fish blood sampling. By the means of a different diet, it was possible to obtain two groups of fish that differ significantly in the permeability of the intestinal epithelium and in the content of D-lactate in the intestine. At the same time, despite the differences between the experimental groups in the content of D-lactate in the intestine and in intestinal permeability, no significant differences in D-lactate level in blood were found between them. Analysis of the composition of the intestinal microbiome by metabarcoding for the 16S rRNA gene revealed the absence of lactobacilli in the production of D-lactate in fish.Results. It was shown that the mechanism of accumulation of D-lactate in the blood plasma in fish is less associated with increased intestinal permeability or hyperproduction of this metabolite by the intestinal microflora and is more associated with the utilization of D-lactate in the body. In the experiment, it was not possible to achieve a significant change in the species composition of the intestinal microflora of trout under the influence of a highcarbohydrate diet for 54 days compared to fish that received high-protein diet. Some tendencies towards changes in the composition of the microflora were found in the contents of the hindgut, and perhaps with a longer exposure, these changes could reach a statistically significant level.
Aim. The aim of the research is to study the symbolic capital of northern cities (semiotic potential) contained in "urban texts" and the influence of the historical memory of a multiethnic region on the political process of the region and the country as a whole, through a comprehensive and interdisciplinary study of the symbolic space of cities in the Republic of Sakha (Yakutia). Methods. The study used the cognitive approach, the method of space-time analysis, and the method of historicism and questionnaire survey. Symbolic space as a text is a heterogeneous content. Results. The results of the study show the importance of a symbolic resource as a brand tool, first, depending on the political context and introduction into the collective memory through a constant "reminder" of its importance and significance in the socio-cultural and everyday life of citizens. The resulting databases of monuments served as the basis for compiling a virtual album of monuments and art objects in the cities of the Republic of Sakha (Yakutia). The research results can practically be applied in the field of education, politics, culture, in the study and promotion of the cultural and historical heritage of the regions of Russia. Conclusions. The urgent problem is the task of preserving the accumulated experience and values, places of memory, relics of the past, as well as ideas about the past that are stored in the memory of its inhabitants and the transfer of this knowledge to the younger generation.
