A new, normal phase high performance liquid chromatography (NP-HPLC) method was developed for separation of Bortezomib (BZB) enantiomers and quantitative determination of (1S,2R)-enantiomer of BZB in active pharmaceutical ingredient (API) samples. The developed method was validated based on International Conference on Harmonisation (ICH) guidelines and it was proved to be accurate, precise and robust. The obtained resolution (RS) between the enantiomers was more than 2. The calibration curve for (1S,2R)-enantiomer was found to be linear in the concentration range of 0.24-5.36 mg/L with regression coefficient (R2) of 0.9998. Additionally, the limit of detection (LOD) and limit of quantification (LOQ) were 0.052 and 0.16 mg/L, respectively. Also, in this study, a precise, sensitive and robust gradient reversed-phase HPLC (RP-HPLC) method was developed and validated for determination of BZB in API samples. The detector response was linear over the concentration range of 0.26-1110.5 mg/L. The values of R2, LOD and LOQ were 0.9999, 0.084 and 0.25 mg/L, respectively. For both NP-HPLC and RP-HPLC methods, all of the RSD (%) values obtained in the precision study were <1.0%. System suitability parameters in terms of tailing factor (TF), number of theoretical plates (N) and RS were TF < 2.0, N > 2,000 and RS > 2.0. The performance of two common integration methods of valley to valley and drop perpendicular for drawing the baseline between two adjacent peaks were investigated for the determination of diastereomeric impurity (Imp-D) in the BZB-API samples. The results showed that the valley to valley method outperform the drop perpendicular method for calculation of Imp-D peak areas. Therefore, valley to valley method was chosen for peak integration.
Objective One of the treatments that have been proposed recently for deaf children is cochlear implant. It is a new technology and an accepted treatment for children with hearing impairment. In recent years, researchers have paid special attention to the family and its relationship with the developmental skills of children with hearing loss. Clinical experience has also shown that emotional performance and psychological problems of the caregiver can have unfavorable effects on cochlear implanted children's developmental skills. Therefore, the present study was conducted with the aim of determining the relationship of parent-child stress with developmental skills of children with cochlear implants. Materials & Methods The present study was a correlational study that was conducted in 2015. The study population comprised all children undergone cochlear implant surgery in Al-Zahra Hospital of Isfahan. Out of them, 57 participants (34 boys and 23 girls) were selected randomly. To evaluate the research variables, the Parenting Stress Index, Leiter International Performance Scale, the Vineland Social Maturity Scale, the Language Development Scale, and the Cochlear Implanted Children's Communication Performance Scale were used. The data of the research were analyzed using descriptive statistics as well as correlational and regression analysis methods. SPSS version 22 was used for data analysis.Results The results of analyzing data indicated that there is a negative (reverse) significant correlation between parent-child stress and language development (-0.59), social development (-0.38), and communication development (-0.54) of children (P≤0.01). According to regression analysis investigating the explanation of language development (criterion variable) based on parent-child stress (predictive variables), the coefficient of determination equals to 0.34 (R 2 =0.34) and thus, 34% of the variance in the dependent variable (language development) are explained via parent-child stress. Also, in explaining the social development (criterion variable) based on parent-child stress (predictive variables), the coefficient of determination equals to 0.14 (R 2 =0.14) and thus, 14% of the variance in the dependent variable (social development) are explained via parent-child stress. Also, in explaining communication function (criterion variable) based on parent-child stress (predictive variables), the coefficient of determination equals to 0.29 (R 2 =0.29) and thus, 29% of the variance in the dependent variable (communication function) are explained via parent-child stress. Conclusion The results of this study showed that parent-child stress had a significant effect on developmental skills of children with cochlear implants. Due to the importance of developmental skills in children with cochlear implants, the results of this study warned the need to provide counseling and psychological support for this group of parents. Introduction: Autism spectrum disorder (ASD) imposes a lot of difficulties and calamities on parents particularly mothers due to close relationship with their children. The present study aimed to examine the effect of transdiagnostic treatment on entrapment and boredom of mothers of children with ASD. Methods: This randomized clinical trial study was conducted on all mothers with ASD children in Isfahan City. Forty participants were initially selected using the convenience sampling method and were randomly divided into two equal groups of control and experimental (n= 20, each group). The transdiagnostic treatment was conducted during 12 sessions of 60 minutes each. Data were collected using the Entrapment Scale and Multidimensional State Boredom Scale.The validity and reliability were measured and confirmed in the previous studies. Data were analyzed by the SPSS 21 software. Results: Results showed a significant difference in internal entrapment (P = 0.001), external entrapment (P = 0.004), disengagement (P = 0.005), high arousal (P = 0.001), low arousal (P = 0.004), inattention (P = 0.010) and time perception (P = 0.022) between the two experimental and control groups in both pre-test and post-test. Conclusions: It seems that the unified transdiagnostic treatment is an appropriate method for reducing psychological problems (such as entrapment and boredom) in mothers with ASD children. Therefore, it is suggested to use this approach with other treatments in educational and treatment centers.
Objectives This study was conducted to design Individual Differences intervention based on parents' expressed emotion and its impact on children's social skills with high-functioning autism disorder. Methods The first part of this study is comparative-causal and the second part is a semi-experimental study. In order to design Floortime treatment plan based on parents' expressed emotion, first in an Embedded Design Mixed Research Method by means of researcher's questionnaire for qualitative measurement and family questionnaire for quantitative part of emotional state of mothers with autistic children were measured. Accordingly, the treatment plan with 23 meetings was codified. Finally, 20 children with high-functioning autism were selected from the autism centers by available sampling. Out of them, 10 children were put into the experimental group and they were provided designed intervention plan in addition to the ABA intervention, and the other 10 children formed the control group, who merely received Applied Behavior Analysis (ABA) intervention. Tools used were Family Questionnaire, ASSQ test, Stanford-Binet intelligence test, and Gilliam and Vineland social compatibility tests. Results Results show that there are significant differences in mother's expressed emotion in case of normal and autistic children. The expressed emotion of mothers of autistic children is high in terms of the total score of expressed emotion, criticism, and Emotional Over-Involvement (EOI). After the intervention, no significant difference was observed in the social compatibility and communication skills between the two groups of children who were under the ABA intervention and who were under Floortime intervention. But after the intervention, the experimental group had a better score in relation variable. Conclusion Given the high level of excitement expressed by parents of children with autism, various interventions have to reduce their excitement. You can also use social interventions such as floortime intervention to increase the association of children with high performance autism disorder.
