Purpose: To investigate the anti-hyperlipidemic effect of Caralluma edulis and Verbena officinalis.Methods: Phytochemical analysis of crude extracts of Caralluma edulis (Ce.Cr) and Verbena officinalis (Vo.Cr) were carried out. Hyperlipidemia was induced in mice with high-fat diet (HFD, 1.25 % w/w cholesterol, 0.5 % w/w cholic acid and 10 % v/w coconut oil). All the groups, except the saline-treated group, were fed on HFD for 4 weeks (lead-in period) to induce hyperlipidemia. Thereafter, the groups were treated with varying doses of the plant extract for 2 weeks (treatment period) as well as atorvastatin (10 mg/kg) reference standard. Body weight was measured fortnightly for all groups. Total cholesterol (TC), triglyceride (TGs) and low density lipoprotein (LDL) were assayed using Merck diagnostic kits. For histopathological analysis, liver slices were fixed in 10 % formalin and embedded in paraffin wax and was examined with the aid of hematoxylin and eosin staining (H & E).Results: Caralluma edulis (Ce.Cr) contains saponins, alkaloids, tannins, phenol, glycosides, terpenoids and flavonoids while Verbena officinalis (Vo.Cr) tested positive for the presence of alkaloids, carbohydrates, flavonoids, saponins and tannins. HFD increased total cholesterol (TC), triglyceride (TGs), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) compared to regulator diet (p < 0.001). Treatment of the animals with Ce.Cr and Vo.Cr dose-dependently (500 - 1000 mg/kg) reduced serum TC, TGs, LDL and VLDL (p < 0.05, p < 0.01, p < 0.001, vs. HFD group) and raised high density lipoprotein (HDL) (p < 0.01, vs. HFD group), similar to that observed with atorvastatin (10 mg/kg). The anti-hyperlipidemic effects of Ce.Cr and Vo.Cr were also confirmed via liver histopathology results, showing improved structure with no hepatocellular necrosis and fat accumulation.Conclusion: These results indicate that Caralluma edulis and Verbena officinalis exhibit antihyperlipidemic effect; thus, the plants have therapeutic potentials for the management of lipid disorders.Keywords: Caralluma edulis, Verbena officinalis, Anti-hyperlipidemia, Hepatocellular necrosis
Idiopathic generalized epilepsy (IGE) is the most prevalent type of epilepsy with genetic origin. Mutations in ion channel genes have been identified as a common cause of IGE. Several studies have reported various epilepsy risk variants of GABRG2 (gamma-aminobutyric acid type A receptor subunit gamma2 subunit) gene in different ethnic groups, but the results are inconsistent. The purpose of this case-control research is to determine if GABRG2 polymorphisms contribute to IGE susceptibility and antiepileptic drug resistance in Pakistani population. For this purpose, we genotyped exon2, exon5 (C540T and C588T), exon7 (T813C), exon8 (K289M), and exon9 of GABRG2 gene by restriction fragment length polymorphism and Sanger’s sequencing in 87 drug-responsive idiopathic generalized epilepsy patients, 55 drug-resistant epilepsy patients, and 83 healthy controls. Restriction fragment length polymorphism (RFLP) and sequencing results indicated only C588T polymorphism in the studied subjects. The comparison of genotypic and allelic frequencies showed significant differences between IGE patients and control groups ( P = 0.008 and odds ratio = 4.2 ) and nonsignificant association of C588T polymorphism in antiseizure medication-resistant patients ( P = 0.9 ). Our findings showed that C588T polymorphism of GABRG2 is a risk variant for IGE in Pakistani population. Further studies are required to validate the results.
The imbalance between modulation of the cell proliferation and apoptosis results in abnormal cell production which ultimately leads to the cancerous condition. During the 21st century, cancer is probably exceeded to infectious and other chronic diseases as the major cause of death in all countries. The increasing global prevalence ratio and a speedily rising population of cancer survivors focus on the ferocity of public health and economic challenges pretended this disease. This review summarizes the significant outcomes of dietary intake which can enhance the immune response and reduce the risk of cancer. The nutritional general mechanism of action to influence the immune function and the potential activity of anticancer immunotherapy are also studied. For several cancers immunotherapy has developed into standard treatment but considerably remains to be done to prolong the activity of its potential efficiency mainly to cancers that are unaffected within each indication. Although a higher consumption of plant foods like vegetables, fruits, legumes, nuts and whole grains has been suggested for cancer prevention, it has been undecided what is the prime amount of ingestion of these foods and whether particular subtypes are valuable. Several million premature mortalities can be prevented if a high intake of plant foods is adopted globally. Furthermore, for reducing the risk of different types of cancers and slow down their development many dietary products have been proven advantageous. Epilepsy is a major global issue. Epilepsy patients are treated with AED (antiepileptic drugs). Interindividual variability in drug response has been documented in several studies. The resistance to drug response may be attributed to genetic polymorphism. The current study was undertaken to investigate the CYP2C9 gene polymorphism associated with antiepileptic drug (AED) resistance in the Pakistani population. The current study included 337 individuals including 100 control subjects, 110 drug-resistant subjects, and 127 drug responders. Genomic DNA was isolated from blood, and amplification of rs1799853 (430C > T) and rs1057910 was carried out by polymerase chain reaction. Genotypes of CYP2C9 SNPs were determined by Sanger’s sequencing. Astounding results were observed in the current study that none of the well-known reported SNPs of CYP2C9 was found in our Pakistani cohorts. However, a novel missense variant (c.374G > A) was found only in drug-resistant patients of the current study. According to the in silico analysis performed by PolyPhen-2, it was observed that this nonsynonymous substitution is likely to be pathogenic. The results of our study demonstrated that rs1799853 and rs1057910 may be involved in drug resistance in the Pakistani population. However, some other variants on CYP2C9 may play a critical role in AED resistance that needs to be explored.
The prevalence of eating disorders among University students has been on rise throughout the world. The present descriptive cross sectional study was conducted to determine the prevalence of eating disorders among the female students of pharmacy department at University of Sargodha, Pakistan using well established EAT-26 questionnaire. The study included 177 female undergraduate pharmacy students from all five years of Pharm D. An EAT-26 score over 20 was indicator of disturbed eating behavior. Out of the total 177 students who participated the study, 47 (26.6%) students had disturbed eating behavior. The mean EAT-26 score was 14.7. According to BMI calculations, 31.64% of total individuals were underweight, 59.32% normal, 6.78% overweight and 2.26% belonged to obese category. The prevalence of eating disorders was found to be much higher than the studies conducted at developed countries. There is an urgent need of an effort to increase awareness of such disorders among students and general public. Conduction of such study at other institutes and at larger scale is recommended.
