Streptococcus suis is a zoonotic agent that causes sepsis and meningitis in pigs and humans. S. suis infections are responsible for large economic losses in pig production. The lack of effective vaccines to prevent the disease has promoted the extensive use of antibiotics worldwide. This has been followed by the emergence of resistance against different classes of antibiotics. The rates of resistance to tetracyclines, lincosamides, and macrolides are extremely high, and resistance has spread worldwide. The genetic origin of S. suis resistance is multiple and includes the production of target-modifying and antibiotic-inactivating enzymes and mutations in antibiotic targets. S. suis genomes contain traits of horizontal gene transfer. Many mobile genetic elements carry a variety of genes that confer resistance to antibiotics as well as genes for autonomous DNA transfer and, thus, S. suis can rapidly acquire multiresistance. In addition, S. suis forms microcolonies on host tissues, which are associations of microorganisms that generate tolerance to antibiotics through a variety of mechanisms and favor the exchange of genetic material. Thus, alternatives to currently used antibiotics are highly demanded. A deep understanding of the mechanisms by which S. suis becomes resistant or tolerant to antibiotics may help to develop novel molecules or combinations of antimicrobials to fight these infections. Meanwhile, phage therapy and vaccination are promising alternative strategies, which could alleviate disease pressure and, thereby, antibiotic use.
Adequate protein levels are necessary to maintain strong honey bee [Apis mellifera (L.)] colonies. The aim of this study was to quantify how pollens with different crude protein contents influence protein stores within individual honey bees. Caged bees were fed one of three diets, consisting of high-protein-content pollen, low-protein-content pollen, or protein-free diet as control; measurements were made based on protein content in hemolymph and fat body, fat body weight, and body weight. Vitellogenin in hemolymph was also measured. Bees fed with high crude protein diet had significantly higher levels of protein in hemolymph and fat bodies. Caged bees did not increase pollen consumption to compensate for the lower protein in the diet, and ingesting approximately 4 mg of protein per bee could achieve levels of 20 microg/microl protein in hemolymph. Worker bees fed with low crude protein diet took more time in reaching similar protein content of the bees that were fed with high crude protein diet. The data showed that fat bodies and body weight were not efficient methods of measuring the protein status of bees. The determination of total protein or vitellogenin concentration in the hemolymph from 13-d-old bees and protein concentration of fat bodies from 9-d-old bees could be good indicators of nutritional status of honey bees.
One of the most important challenges that agriculture faces is sustainable water management and its adaptation to climate change. This adaptation is more important in regions where recurrent draughts and overexploitation of water resources happen. However, historical irrigation systems, such as the Real Acequia de Moncada (RAM) in Valencia, have found innovative approaches to deal with this phenomenon. This paper analyzes the case of Massamagrell and Puçol, which reused the treated waste-water of the closest waste-water treatment plant (WWTP). The study focused from a circular economy perspective on the technological, agronomical, and social implications of this decision. Results show that there are clear benefits for both farmers and WWTP managers. On the one hand, additional nutrients and regularity in their water supply benefit farmers. On the other, WWTP managers can reuse the treated effluent in the system, contributing to the closure of the water cycle and avoiding pumping the treated water into the sea. However, more detailed information and coordination is needed among the different stakeholders. Questions regarding the illegal connection of waste pipes with the traditional irrigation or the payment of pumping costs for reuse have gone unanswered, and there is a need for better reflection from all stakeholders.
Management with synthetic fungicides in the control of phytopathogens in viticulture can cause environmental pollution and contamination with residues in grape cluster. The objective of this study was to verify the effect of aqueous cinnamon extract on the in vitro and in vivo control of Plasmopara viticola on catalase activity on ‘Isabel Precoce’ vines. The treatments used were: aqueous cinnamon extract (ACE) at concentrations of 0.12; 0.25 and 0.50% plus 0.25% vegetable oil (VO); being the standard treatments VO (0.25%), Bordeaux mixture 1:1:100 (lime: copper sulfate: water) and water only. The germination tests of P. viticola sporangia were carried out in incubation periods of 4 and 24 hours of the pathogen in contact with the treatments. In addition, the area under the disease progress curve (AUDPC) and the activity of the catalase enzyme were estimated in plants grown in the greenhouse. The results indicated that the treatments with 0.12%, 0.25% and 0.5% ACE with VO reduced the germination of P. viticola. In relation to the AUDPC, the 0.25% dose of VO associated ACE reduced 65% and 67% in leaf discs and vines in the greenhouse, respectively. This fact is related to the induction of CAT activity provided by this dose in the periods of 2HBA, 2HAI and 4HAI. Thus, it can be said that the ACE associated with VO can be used to control the downy mildew of the ‘Isabel Precoce’ vine.
