SUMMARY Soybean nodulation is a highly controlled process that involves complex gene regulation at both transcriptional and post‐transcriptional levels. In the present study, we profiled gene expression changes, alternative splicing events, and DNA methylation patterns during nodule formation, development, and senescence. The transcriptome data uncovered key transcription patterns of nodule development that included 9669 core genes and 7302 stage‐specific genes. Alternative splicing analysis uncovered a total of 2323 genes that undergo alternative splicing events in at least one nodule developmental stage, with activation of exon skipping and repression of intron retention being the most common splicing events in nodules compared to roots. Approximately 40% of the differentially spliced genes were also differentially expressed at the same nodule developmental stage, implying a substantial association between gene expression and alternative splicing. Genome‐wide‐DNA methylation analysis revealed dynamic changes in nodule methylomes that were specific to each nodule stage, occurred in a sequence‐specific manner, and impacted the expression of 1864 genes. An attractive hypothesis raised by our data is that increased DNA methylation may contribute to the efficiency of alternative splicing. Together, our results provide intriguing insights into the associations between gene expression, alternative splicing, and DNA methylation that may shape transcriptome complexity and proteome specificity in developing soybean nodules.
Bat populations are declining worldwide because of anthropogenic activities, including habitat destruction and hunting. Cambodia represents an important case study for studying human-bat interactions, as loss of karst caves and the destruction of forests threaten the stability of bat populations and the ecosystem services they provide. Cambodians rely on bats for tourism revenue, fertilizer from guano, and as a source of protein. However, there is a lack of information on people's attitudes towards and relationships with cave-roosting bats. In 2018, we interviewed 60 residents around three karst outcrops (Sampeau Hill, Banan Hill, and Reichiatra Hill) in Battambang Province, northwestern Cambodia, along with agricultural professionals in Battambang Town, the province's capital. The primary objectives of the interviews were to examine people's (1) attitudes towards bats, (2) experiences with bats, and (3) engagement in high-risk behaviors associated with transmission of bat-related diseases (e.g., guano mining, hunting, etc.). Most respondents (70%) held positive attitudes towards bats and listed guano production, pest control, and tourism as benefits bats provide. Additionally, all informants believed bats should be protected and stated that they would feel sad if bats were extirpated. Conversely, respondents noted that many people eat bats. We followed these semi-structured interviews with five key informant interviews involved with the conservation of bats, which provided information on the history of human-bat interactions within these communities. Respondents' positive attitudes towards bats and recognition of ecosystem services bats provide indicate they would support bat conservation policy and may be interested in developing community-based conservation programs around karst outcrops.
The number of the world's food insecure rose at the end of the first decade of the twenty-first century. Despite these negative developments, however, a 2010 United Nations report argues that food security could be improved if development efforts are supported by government programs that target smallholder farmers. This report is significant because it challenges the neoliberal perspective, which tends to promote a private-enterprise market system and favor large-scale producers. These competing visions for agricultural development frame our evaluation of the impact of the Africa Rice Center's (AfricaRice) efforts to promote new rice varieties among smallholder farmers to narrow the rice consumption-production gap in Ghana. We begin by distinguishing the outreach efforts to help farmers increase production and the political-economic conditions that limit the longevity of that outreach effort. We reviewed program documents revealing expenditures and yields among the smallholder farmers, and we conducted intensive interviews with the farmers. Although we find that the program succeeded in mobilizing farmers to increase rice production, we question the sustainability of the program's impact because state funding for the program has ended.
Within the last decade, Sustainable Intensification (SI) has emerged as a strategy to respond to future food security challenges. It incorporates increased food production without the cultivation of more land while incurring no net environmental cost. Frameworks attempting to measure SI often focus heavily on production indicators while overlooking other important indicators, such as impacts on economic, social, or human conditions. In this study, we evaluate the purposeful assemblage and management of neglected and underutilized species (NUS) in fringe areas around rural homesteads as a potential SI strategy. We use a recent SI assessment framework developed by the Feed the Future Innovation Lab for Collaborative Research on Sustainable Intensification (SIIL) that incorporates five measurable domains (productivity, economic, environmental, human condition, and social). We present findings from a qualitative case study in northwest Cambodia to assess local conceptualization of wild gardening, current uses of NUS, perceived benefits and challenges to their use and management, and the potential of wild gardening as a SI strategy. The qualitative methods employed 65 key informant interviews and four focus group discussions with both men and women participants. Our results indicate that wild gardening is an important component of rural livelihoods in northeast Cambodia. However, a general lack of knowledge of strategic benefits, such as nutrition potential, inhibits its use for maximum benefits. Wild gardening addresses multiple SI domains simultaneously and demonstrates the potential to be a promising SI strategy for improving rural livelihoods in Cambodia.
