Underwater drilling blasting technique is often adopted to clear away the obstructed reef and dredge-cut in the mountainous waterway dredging engineering. However, the vibration of the underwater blasting and excavation of the riverbed has a certain in uence on the stability of bank slopes. Combined with the partial bank collapse phenomenon in the waterway regulation project between two dams in the Three Gorges reservoir area, the change of pore water pressure of unsaturated soil under heavy rainfall and the increase of permanent displacement of soil under blasting vibration in the reservoir area are discussed. This paper explains the weakening effect of rainfall in ltration and blasting vibration on the stability of the bank slope. By studying the law of slope failure deformation under the separate action and coupling action, it is found that rainfall in ltration promotes the increase of slope permanent displacement and the blasting vibration enlarges the in ltration area of the soil, which demonstrated the accelerated failure mechanism of the rainfall-blasting slope.
Decreased expression of Src homology region 2 domain containing protein tyrosine phosphatase 1 (SHP-1) was found in human breast cancer cell lines and primary breast cancers. As a candidate tumor suppressor gene, the role of SHP-1 in HER-2-positive breast cancer remains to be elucidated. The aim of the present study was to investigate the effects of upregulated SHP-1 expression in reversing trastuzumab resistance and migration by HER-2-positive breast cancer cells, potentially providing a new target to trastuzumab resistance in anti-HER2 therapy. Western blotting analysis was used to determine the expression levels of SHP-1 in trastuzumab-resistant cells transfected with a lentivirus vector expressing GFP-(control) or GFP-SHP-1 (experimental). MTT assay was used to determine changes in sensitivity to trastuzumab between two modified cell lines after overexpressing SHP-1. Clone formation and wound-healing assays were used to detect the capacity change in cell proliferation and migration. Western blot analysis was also used to examine the expressions of c-myc, cyclinD1, p-STAT3, STAT3, and MMP-9. Results from this study indicated that SHP-1 not only reduced cell proliferation and migration, but also significantly sensitized resistant cells to trastuzumab. Data also showed that SHP-1 restores sensitivity to trastuzumab in drug-resistant cell lines. Mechanistic analyses indicated that SHP-1 might target STAT3 to inactivate p-STAT3 and reduce expression of its downstream signaling pathway. In conclusion, results presented here suggest that targeting SHP-1 may represent a potential approach to re-sensitize previously resistant cells to trastuzumab treatment.
The existing tunnel lining structure is easily affected by geological conditions, disturbance in the early stage of construction and operation in the later stage. This paper analyzes the main factors affecting the lining structure diseases of existing tunnels from three aspects of engineering construction, natural environment and engineering geology, starting from 13 independent factors affecting the lining structure diseases of existing tunnels. First of all, the index system of the formation factors of lining diseases in the reconstruction and expansion tunnel is established by using the analytic hierarchy process (AHP), and the hierarchical index system is put forward. Based on AHP-Extenics model, the evaluation method of lining structure disease of existing tunnel is proposed. Then, combined with engineering practice and expert scoring, the weight of safety evaluation index is determined, and the safety evaluation index and weight are expanded by using correlation function. Finally, the disease evaluation grade of the existing tunnel lining structure is obtained. This method uses the formal Extenics comprehensive evaluation model, which makes the calculation results more objective and provides a new method for the disease evaluation of the existing tunnel lining structure.
